Experimental Study On Effect Of Acupuncture At "Feishu" Point On M1M2M3 In Rats With Lung Immune Inflammation

Purpose : After electro acupunctured at "Feishu"(BL 13) points of lung immune inflammation model rats, to observe the changes of total number of inflammatory cells and the total number of eosinophils(EOS) in the bronchoalveolar lavage fluid(BALF),morphologic changes of lung, the changes of relative protein content and expression of m RNA of parasympathetic muscarinic receptors M1, M2, M3 in lung tissue, to discuss the mechanism of parasympathetic nerve between "Feishu"(BL 13) point and the lung.Materials and methods:40 healthy male Wistar rats were randomly divided into normal group, model group, "Feishu"(BL 13) group, non acupoint group,10 rats in each group, according to the random number table method. The model of allergic airway immune was made by injecting the mixture of aluminum hydroxide(Al(OH) 3)and ovalbumin(OVA)into abdominal(0, 7, 14 and 21 days),and inhalating nebulized ova(22-43 days, once every two days, 20 minutes every time).The acupuncture group chosed "Feishu"(BL 13) which are on both sides of the spine(third thoracic intercostal under both sides) according to“Experimental acupuncture and moxibustion”, and was electro acupunctured into about3mm(density wave). The intensity of acupuncture was the needle handle trembling, the acupuncture time is 20 minutes, once every other day, a total of 3 weeks. The non acupoint group selected points next to "Feishu"(BL 13) about 5mm and the treatment method and the time were consistent with the above. The relative contents and the expression of m RNA of M1, M2 and M3 were detected by Western blot and Real-time PCR.Results:1 The total number of inflammatory cells and EOS in BALF Compared with the model group, the total number of inflammatory cells and EOS in the normal group and "Feishu"(BL 13) group were significantly decreased(P <0.01), in non acupoint group the total number of inflammatory cells were significantly lower(P<0.05),and the total number of EOS was significantly decreased(P < 0.01); Compared with the normal group, the total number of inflammatory cells and EOS in "Feishu"(BL 13) group were significantly increased(P<0.05); the total number of inflammatory cells and EOS in non acupoint group were significantly increased(P<0.01).2 The changes of lung tissue and tracheal morphology In model group, the airway epithelium of rats were off and necrosis and airway smooth muscle were thickened. There was obvious congestion and edema in the mucosa.The lumen became narrow. A large number of inflammatory cells infiltrated into the lumen and the airway mucosa. Compared with the model group, the phenomenon of congestion and edema and inflammatory cell infiltration were not so obvious in "Feishu"(BL 13) group, but compared with the normal group, it was more obvious.3 The expression leve of M1, M2 and M3 m RNA in lung tissues Compared with the normal group, M1 m RNA and M3 m RNA were significantly higher,M2 m RNA was decreased significantly in model group(M1 m RNA P<0.05, M2 m RNA P<0.01, M3 m RNA P<0.01); Compared with the model group, the M1 m RNA and M3 m RNA were decreased significantly, M2 m RNA was significantly higher in "Feishu"(BL 13)group(M1 m RNA P<0.05, M2 m RNA P<0.01, M3 m RNA P<0.01); Compared with the normal group, M2 m RNA was decreased significantly, M3 m RNA was increased significantly, and M1 m RNA was not statistically significant in non acupoint group.(M1m RNA P>0.05, M2 m RNA P<0.01, M3 m RNA P<0.05).4 The relative contents of M1, M2 and M3 receptor protein in lung tissues Compared with the normal group, M1 and M3 were significantly higher and M2 was significantly decreased in model group( M1 P<0.01, M2 P<0.01, M3 P<0.05);Compared with the model group, M1 and M3 were decreased significantly and M2 was significantly higher in "Feishu"(BL 13) group( M1 P<0.01, M2 P<0.05, M3 P<0.05); Compared with the normal group, the M1 was increased significantly, M2 was significantly lower, and M3 was not statistically significant in non acupoint group.( M1 P<0.01, M2 P<0.05, M3 P>0.05).Conclusion:1 “Feishu”(BL 13) can effectively reduce the total number of inflammatory cells and EOS in BALF of lung immune inflammation rats.2 The expression of M1 and M3 was increased and the expression of M2 was decreased in the lung tissues of the rats with pulmonary inflammation.3 “Feishu”(BL 13) can effectively reduce the expression of M1 and M3 and increase expression of M2 in the lung tissues of the rats with pulmonary inflammation.4 “Feishu”(BL 13) can protective M2 receptor.5 “Feishu”(BL 13) can regulate the pathological changes of lung by the parasympathetic.
