Objective:Air pollution is a widely watched environmental issue currently.Particles less than 2.5 micrometers in diameter,known as PM2.5 has been proved to be an important pathogenic of many different kind of disease,especially lung disease.PM2.5 particles can invade the respiratory system directly, cause inflammatory reaction and chronic damages in lung and trachea.PM2.5 can also induce or aggravate multiple system diseases.A large number of studies has shown that traditional Chinese medicine could be helpful for chronic obstructive pulmonary disease,asthma and other respiratory diseases.Our former experiments has proved that Gubenzhike formula can improve the chronic pulmonary injuries.Based on strengthening body resistance, Gubenzhike formula has effects on both temporary and permanent symptoms. Gubenzhike formula intented to release the damage to respiratory system caused by PM2.5 by improving the body’s immune and defence function.By establishing the PM2.5-induced mouse model with lung injury and measuring the expression of mouse lung tissue IL-8, TNF-a and SIgA,this research is going to do further studies of the damage mechanism of PM2.5 to respiratory system,including the influence and effect to inflammatory response,the regulation of inflammatory factors and the body’s autoimmunization. It will provide more experimental bases for prevention and treatment of respiratory diseases by using Gubenzhike formula.Methods:PM2.5-induced mouse models with lung injury were established by nasal dripping of PM2.5 suspension liquid,and the traditional Chinese medicine intervention group was built up by gavage of Gubenzhike solution.The mice were divided into 3 groups:blank control,lung injury model and traditional Chinese medicine intervention group.Determined expression of IL-8 and SIgA in lung homogenates by PCR,and determined TNF level with ELISA.The experimental data were analyzed by SPSS22.0.Results:The PM2.5-induced models group shows obvious inflammatory cell infiltration in the lung tissue,mainly with neutrophils and lymphocyte.The the alveolar septa was widen and the alveolar space increased.The bronchial mucosa and bronchial wall were damaged in varying degrees.In the traditional Chinese medicine intervention group,the inflammatory infiltration in lung tissue were decreased,and the structure of bronchiole and pulmonary alveolus was more complete.The content of IL-8,TNF-α and SIgA in lung tissue of PM2.5-induced mouse models was significantly higher than that of the blank control group.Levels of IL-8,TNF-a and SIgA in Chinese medicine intervention group were lower than that of The PM2.5-induced models group.The expression levels of IL-8 and TNF-a in Chinese medicine intervention group were higher than that of the blank control group,and the SigA level was quite close to that of blank control group.Conclusion:1.According to the morphological changes of lung tissue,Gubenzhike formula can reduce the inflammatory response which caused by PM2.5,and reduce inflammatory infiltrates in the lung,it also can promote the rehabilitation of lung tissue.2. Gubenzhike formula can reduce the secretion of IL-8 and TNF-a in the lung tissue of PM2.5-induced models. Gubenzhike formula can reduce respiratory chronic inflammation by inhibiting the expression of inflammatory cell factors.3. Gubenzhike formula can regulate the secretion of SIgA. Gubenzhike formula reduced SIgA in lung tissue of PM2.5-induced mouse models to normal level,which helps to balance the immune system and improve the body’s self regulation mechanism.
