Protection Effect Of Zingiber Extract On Rat Retinal Ischemia-reperfusion Injury

Objective: To investigate the protection effect and mechanism of Zingiber extract on rat retinal ischemia-reperfusion injuryMethods: The forty five SD rats were divided randomly into three groups: control group, ischemia group and treatment group. RIR models were made with infusing normal saline to anterior chanlbers and increasing pressure for l hour. The treatment group were given Zingiber extract and distilled water mixture 400mg?kg-1?d-1 by intragastric administration, The control and the ischemia group were given distilled water simultaneously, twice a day for five days before making RIR models. At 12 hour, 24 hour, 48 hour and 72 hour after the ischemia-reperfusion damage, the content of the superoxide dismutase (SOD), malonyldialdehyde(MDA) and nitric oxide(NO) were measured by colorimetric method, the retinal ultrastructural changes were detected under the electron microscope.Results: 1. Compared with control group, retinal MDA,NO content increased and SOD vitality reduced in ischemia group, there was statistically significant difference (P<0.05); Compared with ischemia group, retinal MDA,NO content reduced and SOD vitality increased in treatment group every time point, there was statistically significant difference (P<0.05); Compared with control group, retinal SOD vitality reduced in treatment group, but there was not statistically significant difference at 12h(P>0.05); The MDA content increased, there was statistically significant difference (P<0.05); The NO content increased , but no statistically significant difference at 12h,72h(P>0.05). 2. In ischemia group, RGCs apoptotic cells were seen and there were lot's of myelinfigures because of cellular membrane destroyed. Among retinal nerve fibre layer, cells were swollen and kytoplasm was vacuolization; cytochondriomes electron density cutted down, cristae disrupted and confused; acromere was disorganization, and arrangement became disordered, the number decreased under the electronmicroscope. In treatment group, cells'morphous kept fairly integrity, swelling abated, myelinfigures reduced ,kytoplasm electron density was uniformity, cristae disrupt wasn't manifestation, acromere arranged in order and the number was more than ischemia group.Conclusion: 1. We could successfully made experimental RIRI model by infusing normal saline to anterior chanlbers and it could be certified under the electronmicroscope.2. Our study showed that, zingiber extract could protect retinal from ischemia-reperfusion injury by reducing retinal MDA, NO content, improving SOD vitality and palliating the ultrastructure damage.
