| Objective Prepare the model of retinal ischemia-reperfusion performed in the method of thread embolism,and investigate the mechanism of protection that Neuregulin-1βpreconditioning on retinal ischemia-reperfusion injury in rats.Methods Mirror the method of thread embolism to prepare the model of cerebral ischemia-reperfusion,and prepare the model of retinal ischemia-reperfusion.The rats in the group of preconditioning receive intraocular injection of Neuregulin-1β15μat 24h before retinal ischemia. ischemia last 1h,then resume the blood supply of the retina.Rats were grouped and sacrificed respective at1h,6h,12h,24h,48h and 72h.then eyes were enucleated.Retinal paraffin sections of rats were stained with TdT-mediated dUTP nick end labeling(TUNEL)to examine apoptosis of gangliocyte and obtain statistics conclusions,meanwhile detect the Bcl-2 and Bax expression in the method of immunohistochemistry.Result the apoptosis of gangliocyte emerge at 1h after reperfusion,increasing progressively,and it culminates at 24h after reperfusion,then it decrease at 48h after reperfusion,the apoptosis still exists at 72h after reperfusion.Compared to the ischemia-reperfusion group,the Neuregulin-1βgroups exhibited an decrease apoptosis of gangliocyte after reperfusion every time point of retina ischemia (all P＜0.01).Compared to the ischemia-reperfusion group,The Neuregulin-1βpretreated group exhibited a significant increase of Bcl-2 expression and reduction of Bax expression after reperfusion every time point of retina ischemia(all P＜0.01).Conclusions The model of retinal ischemia-reperfusion can be performed successfully in the method of thread embolism,The phenomenon of apoptosis is obvious in the inner retina after ischemia-reperfusion injury,especially in the group of RIR24h.The mechanism that inhibit the apoptosis of gangliocyte induced by Neuregulin-1βpreconditioning may be related to up-regulation of Bcl-2 exoression and down-regulation of Bax expression. |
