Protection Effect Of Minocycline On Rat Retinal Ischemia/Reperfusion Injury

Objective Retinal ischemia/reperfusion injury is a common eye disease, mainly occurrs in acute angle closure glaucoma, the central dynamic of retinal vein thrombosis, diabetic retinopathy and other retinal vascular obstruction caused by ischemic disease. After blood Recanalization again later,many patients with ischemic oculopathy get serious retinal damage and a further decline in visual function. Retinal ischemia/reperfusion injury is the result of many factors, including oxygen free radicals damage,cells within the calcium overload phenomenon, the role of leukocyte and apoptosis. In this study, we acutely increased the intraocular pressure of rats by infusing anterior chamber to establish the retinal ischemia/reperfusion model.We approached the mechanism and neuroprotective effect of minocycline in the rat retinal ischemia/reperfusion injury. So we can offer preliminary experiment basis to find a safe and effective neuroprotective drug for Clinical use.Methods Adult and healthy SD rats were divided into normal group, ischemia/reperfusion group, treatment group and the sham group.We used the 41/2 scalp needle to infuse the anterior chamber one hour to approached the model,and the infusion bottle height was 150cm.Rats in the treatment group intraperitoneal injected minocycline, rats in the normal group,the sham group and ischemic reperfusion group intraperitoneal injected saline part. aeq.. Reperfusion 6 h, 24h, 48h and 72 h later, retinal content of superoxide dismutase (superoxide dismutase, SOD), MDA (maleic dialdehyde, MDA), nitric oxide (NO) content of were detected by spectrophotometer. Immunization group of semi-quantitative detection of retinal Caspase-3 expression was detected by Immune- histochemistry,and the retinal ultrastructure was observed by transmission electron microscope.Results Compared with the normal retina, retinal SOD content in ischemia/reperfusion group decreased , with MDA and NO content increased;Caspase-3 expression levels increased,with RGC karyopyknosis, shape collapsed.And the membrane is not complete, the nuclear chromatin marginated with apoptotic body. Membranous disc structure was destroyed with myelinfigure. nerve fibers mitochondrial swelled and breaked down with vacuoles. Compared with ischemic/reperfusion group, retinal SOD in the treatment group increased, and MDA,NO decreased;Caspase-3 expression got lower, retinal ultrastructure damage lessened. There was no significant difference between the sham group and the normal group.Conclusions We successfully established a rat retinal ischemia/reperfusion model and can meet the further experiment needs. For retinal ischemia/reperfusion injury, minocycline can increase SOD content, degrade MDA and NO content,improve retina ultrastructure to protect the retina.
