Objective:Investigate the situation of human papillomavirus infection and the distribution of HPV genotype in Chongqing, and observe the association of HPV type and the HPV multi-infection with the level of cervical lesions. Discuss the clinical practice value of gene chip test.Methods: 360 cervical exfoliated cyte samples were collected to detect with gene chip test and thinprep cytology test, and classify the cervical squamous intraepithelial lesion according TCT results. DNA sequencing were used to testify the detective ability of HPV gene chip test and HC2.Results:The HPV positive rate was 42.5%, thereinto the high-risk and low-risk HPV infection rates were 34.17% and 15.56%. The multi-infection rate was 15.00%. 27 types of HPV were detected, and the three most prevalent types in Chongqing were HPV16, HPV58 and HPV52 in order. The high-risk HPV infection rate was increasing with the aggravation of the cervical squamous intraepithelial lesion; there were significant differences between any two groups. The multi-infection rates and virus load were different between normal and abnormal group, but no statistical difference among different levels of cervical lesion. The infection rates of HPV16, 33, 52, 58 in abnormal group were significantly higher than that in normal group. The sensitivity, specificity and Youden's index of gene chip test and Hybrid Capture 2 were 98.33%, 93.40%, 0.917 and 97.56%, 98.17%, 0.957 respectively.Conclusions:The three most prevalent types in Chongqing were HPV16, HPV58 and HPV52.High risk human papillomavirus infection was the main factor of cervical lesion. As the aggravation of the cervical squamous intraepithelial lesion, the infective rate of HR-HPV was significantly increasing. HPV multi-infection made sense to the cervical squamous intraepithelial lesion, but nonsense to the level of lesion. HPV16, 33, 52, 58 were more dangerous to cervical lesion. Gene chip test had high sensitivity specificity and good consistency with Hybrid Capture 2, also can tell the HPV type, so it could be used in the clinical cervical cancer screening.
