Objiect Chinese Drug RHIZOMA DIPHYLLEIAE was called Guijiu, Bajiaopan, Mamudugong in Chinese ancient herbals. There were many reports about people who got to be paralyzed or died for taking RHIZOMA DIPHYLLEIAE overdose. Emphasize the study about quality criteria was to promote safer and more effective clinical medication, and to decrease the waste of. natural resources of Chinese medicinal materials.Methods 1. Collectted the plant specimen of RHIZOMA DIPHYLLEIAE, then identified its botanical origin through appearance, characters, microscopic, physical and chemical identification identification.2. Analyzed finger printing about 10 batchs of 3 different RHIZOMA DIPHYLLEI-AE with the method of HPLC, taking the peak of Podophyllotoxin in RHIZOMA DIPHYLLEIA-E as a referrence. The column was DiamonsilC18 (5μm,250mm×4.6mm), the mobile phase was the solution of 0.1M dihydrogen phosphate buffer:methanol (45: 55). The UV detection wavelength was 290nm. The column temperature was 30℃. And the flow rate was 1.0mL·min-13. Analyzed the dosage, clinical remedy effect of 46 intoxicated cases with RHIZOMA DIPHYLLEIAE by oral administration to approach the safe dose.Results 1. Established the three RHIZOMA DIPHYLLEIAE with different botanical origin, which were oringinated from Dysosma veraipellis (Hance.) M. Cheng, D. pleiantha (Hance.) Woods. And Diphylleia sinensis H. L. Li., by studying the identification, moisture content measure, assaying, property, flavor, meridian distribution, indication, administration, dosage, attention and storage. And which were in Hubei province Chinese Drug quality criteria.2. There were 4 same peaks as the basic characteristic absorption band in the finger prints about 10 batchs of 3 different botanical origins, and there were better similarity, if the botanical origins were the same.3. Toxic dosage was different, some were got intoxication with 0.4g, some were got intoxication with more than 4g.Conclusion 1. Chinese Drug RHIZOMA DIPHYLLEIAE was called Guijiu in Chinese ancient herbals. Chinese Drug was the dry rhizoma of Podophyllum emodi Wall., Podophyllum emodii Wall. var. chinense Sprague, Diphylleia sinensis Li., Dysosma veraipellis (Hance.) M. Cheng, or D. pleiantha (Hance.) Woods.. And that the main'botanical origins of RHIZOMA DIPHYLLEIAE were Diphylleia sinensis Li. Dysosma veraipellis (Hance.) M. Cheng, and D. pleiantha (Hance.) Woods..2. Through the analysis fingerprint about 10 batchs of 3 different RHIZOMA DIPHYLLEIAE with the method of HPLC, it could indicate that no matter the habitat and botanical origin, they all had 4 common peaks when made Podophyllotoxin as the index, besides that, closer genetic relationship made more common peaks.3. Podophyllum and RHIZOMA DIPHYLLEIAE were named by the forma, so it was easy to misuse them. The crude drug should be named by the botanocal oringin, they were Podophyllum emodii Wall. var. chinense Sprague, Diphylleia sinensis Li., Dysosma veraipellis (Hance.) M. Cheng (including D. pleiantha (Hance.) Woods.), and they also should be administrated as poisonous drug. They all should be for external use usually. For oral use, the dosage should be controled below 3g with the doctor's indication. And generally speaking, they should not be in broth, and better be used below 3 days. RHIZOMA DIPHYLLEIAE was better for external use than for oral use, and the dosage should be controlled between 1.5g and 3.0g. And it should not be took more than 3 days.
