Research On Antihypertensive Principals And Quality Analysis Of Radix Et Rhizoma Ephedrae

Radix et Rhizoma Ephedrae,the drying root and rhizome of Ephedra sinica Stapf or Ephedra intermedia Schrenk et C.A.Mey.which belongs to the Ephedrae genus of Ephedraceae.Radix et Rhizoma Ephedrae is embodied in Pharmacopoeia of People's Republic of China(2005 edition). Radix et Rhizoma Ephedrae has the efficacy of stopping sweating and used as antiperspirant in clinic.Modern pharmacological researches have shown that Radix et Rhizoma Ephedrae has significant effect on antihypertension.The dissertation researched on its material foundation and quality evaluation system of antihypertension in order toprovide scientific basis on exploitation and utilization of the medicinal plant.Radix et Rhizoma Ephedrae appropriately smashed was extracted using 85%ethanol,The extraction using the method of systematic solvent extract by petroleum,ethyl acetate and n-Butanol respectively,then got the petroleum ether fraction,ethyl acetate fraction,n-Butanol fraction and so on. The screening of these fractions based on antihypertension was carried out on spontaneously hypertensive rat by the whole animal experiment.The results showed that the fraction of ethyl acetate and n-Butanol had significant effect on antihypertension activity.Therefore,the fraction of ethyl acetate and n-Butanol are activity fraction of Radix et Rhizoma Ephedrae on antihypertension which establish a foundation for the research of the active components of antihypertension. 10 compounds from the ethyl acetate fraction were isolated by various kinds of chromatographies and elucidated 7 of them.Their structures have been determined as Epiafzelechin,β-sitosterol,Stigmasterol-3-O-β-D-glucopyranoside,Mahuannin D,Mahuannin E,Ephedrannin and Ethyl Caprylate.Epiafzelechin,β-sitosterol,Stigmasterol-3-O-D-glucopyranoside and Ethyl Caprylate were found from Radix et Rhizoma Ephedrae for the first time.The structures of compounds Bb,Bf,DeCa-A are being further determined.According to the antihypertension,the five compounds separated from the fraction of ethyl acetate were screened on the SHR by means of whole animal experiment.The results demonstrated that Epiafzelechin and Ephedrannin had good effect on anti-hypertension on spontaneously hypertensive rat,in addition Epiafzelechin was the first discoveried active components of antihypertension from the Radix et Rhizoma Ephedrae. Therefore,it would be scientific basis for annotating the material foundation of Radix et Rhizoma Ephedrae for antihypertension and well foundation for the further research.The character,micro-appreciation,TLC,moisture and heavy metal check and Ephedrannin content measure of Radix et Rhizoma Ephedrae were investigated systemly adopting the modern analytical method.The perfect quality standards was also established.Thus it provides some scientific basis for evaluating the quality and medicating the safety of Radix et Rhizoma Ephedrae.
