Objective: Observing the influence of body acupuncture and high-frequency electroacupuncture on hypothalamic obesity rats' Lee's exponent, TG, TC, LDL-C, HDL-C, leptin, Ins, NPY through the animal experiment. Studying the effect of these two methods on losing weight and discussing their mechanism and differences,then to provide experimental bases for acupuncture-moxibustion clinical treatment of simple obesity.Methods:Duplicated rat hypothalamic obesity model by injecting L-MSG under the skin of neck. Selected weight 7~12g of healthy SD rats 6 nests(male and female),and randomly choosed 8~10 rats from per nest of SD that were born one day. Hypodermic injected 15% normal saline solution of L-MSG at 0.2ml/10g everyday for 5 days under the skin of neck,at the same time the remaining rats were injected the same dosage of physiologic saline as a control.21-day-old about weaning, the male and female divide the cage to raise,the L-MSG-treated rats were fed high lipid bait vessel,without the L-MSG-treated rats were fed ordinary rat bait vessel.36-day-old modeling end,all rats were measured body weight and length,and then calculated Lee's exponent[体重(g)1/3×103/体长(cm)],Lee's exponent higher than normal over 30 for the successful modeling,those didn't meet the criterion were given up, and 30 obesity rats were obtained that were randomly divided into 3 groups,named high-frequency electroacupuncture group(referred to as"electroacupuncture group"(10 rats), body acupuncture group(10 rats), Pathological model group (referred to as"model group"10 rats), and normal control group(injected physiologic saline 10 rats). Electroacupuncture group fixed in a self-control rat machine,were not anaesthetized,selected bilateral "Housanli","Sanyinjiao","Qiuchi","Fenglong","Guanyuan"and"Zhongwan",which were fellowed by switching WQ1002F-type electroacupuncture instrument with rarefaction wave(100Hz), and then acupunctured at 30min/time·d-1 for 28 days.Body acupuncture group used the same fixture method,and adopt normal reinforcement and normal reduction(twist angle 270°,twist frequency 100 times/min)after inserting in the above-mentioned points,then travelled needle 1 min and 1 time/5 min at 30min/time·d-1 for 28 days. Model group and normal control group fixed in the same way at 30min/time·d-1,but did not impose other interventions. Other three groups were fed by high lipid bait vessel besides control group by ordinary rat bait vessel from Hebei Medical University experimental animal center during the acupuncture intervention.Four groups were measured body weight,length by special messenger after intervention,and calculated Lee's exponent.Next day,blood was obtained from arteria femoralis,then examined TG,TC,LDL-C,HDL-C,Ins,leptin,NPY of blood plasm,leptin,NPY of hypothalamus,and observed the pathomorphological changes of rat adipose cell at the same time.All the data were made statistics analysis with SPSS13.0 software.Result:1 Lee's exponent after modelingAfter modeling,Lee's exponent of model group,electroacupuncture group,body acupuncture group(338.02±12.32,349.21±11.61,344.82±18.78)were significantly higher than normal control group(292.66±7.13)more than 30(P<0.01),show that the model successfully.Model group,electroacupuncture group and body acupuncture group had no significant difference in the Lee's exponent(P>0.05),which showed that there was no marked difference between three groups before acupuncturing.2 Impact on obese rats' body weight,length and Lee's exponent of different acupuncture methods2.1 Impact on the obese rats' Lee's exponent before and after acupuncture intervention After intervening for 4 weeks in acupuncture,Lee's exponent of body acupuncture group and electroacupuncture group(306.89±11.23,317.38±15.95)were lower significantly than before(344.82±18.78,349.21±11.61),Prove that the acupuncture treatment is effective.2.2 Impact on the obese rats' Lee's exponent after acupuncture interventionAfter intervening for 4 weeks in acupuncture, Lee's exponent of model group(331.78±5.80)was significantly higher than normal control group(305.41±5.20),the difference has statistics meaning (P<0.01);Lee's exponent of body acupuncture group and electroacupuncture group (306.89±11.23,317.38±15.95) more significantly decreased than model group(P<0.01),which proved that acupuncture could achieve the goal of losing weight;the body acupuncture group was lower than the electroacupuncture group(P<0.05),the body acupuncture group was superior to the electroacupuncture group in reducing Lee's exponent.3 Effect of different acupuncture methods on perirenal fat weight,pericardial fat weight,epiploon adipose weight of obese ratsAfter acupuncture intervention for 4 weeks,perirenal fat weight,pericardial fat weight,epiploon adipose weight of model(2.06±0.48,0.42±0.07,2.85±0.61)mg were higher significantly than normal control group(0.58±0.19,0.23±0.06,1.29±0.29)mg (P<0.01);electroacupuncture group and body acupuncture group were lower significantly than model group(P<0.01 or P<0.05),the electroacupuncture group was superior to the body acupuncture group in reducing obese rats' fat weight.4 Effect of different acupuncture methods on lipid metabolism of obese rats4.1 Effect of different acupuncture methods on serum TG of obese ratsAfter acupuncture intervention for 4 weeks,serum TG of model group(2.42±0.40)mmol/L was higher significantly than normal control group(1.02±0.32)mmol/L (P<0.01),serum TG of electroacupuncture group and body acupuncture group(1.42±0.33,1.81±0.41)mmol/L were lower obviously than model group(P<0.01).The electroacupuncture group was lower than body acupuncture group(P<0.05).4.2 Effect of different acupuncture methods on serum TC of obese ratsAfter intervening for 4 weeks in acupuncture,serum TC of model group(3.39±0.27)mmol/L was higher significantly than normal control group(2.17±0.10)mmol/L (P<0.01),serum TC of electroacupuncture group and body acupuncture group(2.61±0.28,3.05±0.43)mmol/L were lower obviously than model group(P<0.01 or P<0.05).The electroacupuncture group was lower obviously than body acupuncture group (P<0.01).4.3 Effect of different acupuncture methods on serum LDL-C of obese ratsAfter intervening for 4 weeks in acupuncture,serum LDL-C of model group(1.42±0.25)mmol/L was higher significantly than normal control group(1.01±0.19)mmol/L (P<0.01),serum LDL-C of electroacupuncture group and body acupuncture group(1.20±0.13,1.23±0.21)mmol/L were lower obviously than model group(P<0.05).There was no significant difference in the content of serum LDL-C between electroacupuncture group and body acupuncture group(P>0.05),but body acupuncture group,electroacupuncture group had the tendency to decrease progressively in reducing serum LDL-C.4.4 Effect of different acupuncture methods on serum HDL-C of obese ratsAfter acupuncture intervention for 4 weeks,serum HDL-C of model group(0.45±0.08)mmol/L was lower significantly than normal control group(1.03±0.16)mmol/L (P<0.01), serum HDL-C of electroacupuncture group and body acupuncture group(0.57±0.08,0.69±0.14)mmol/L gave up in various degree comparing with model group(P<0.01 or P<0.05).But the body acupuncture group recovered significantly,comparing with electroacupuncture group (P<0.05).5 Effect of different acupuncture methods on serum Ins of obese ratsAfter acupuncture intervention for 4 weeks,serum Ins of model group(38.06±5.89)uIU/ml was higher significantly than normal control group(25.06±2.95)uIU/ml(P<0.01),serum Ins of electroacupuncture group(30.20±4.88)uIU/ml was lower obviously than model group(P<0.01),but there was no significant difference in the content of serum Ins between body acupuncture group(36.63±4.72)uIU/ml and model group,the electroacupuncture group was superior to the body acupuncture group with significance(P<0.01).6 Effect of different acupuncture methods on serum leptin and hypothalamic leptin of obese rats6.1 Effect of different acupuncture methods on serum leptin of obese ratsAfter intervening for 4 weeks in acupuncture,serum leptin of model group(2.58±0.23)ng/ml was higher significantly than normal control group(1.56±0.18)ng/ml (P<0.01),serum leptin of body acupuncture group and electroacupuncture group(1.83±0.19,2.06±0.14)ng/ml were lower obviously than model group(P<0.01).The body acupuncture group was superior to the electroacupuncture group with significance(P<0.01).6.2 Effect of different acupuncture methods on hypothalamic leptin of obese rats.After intervening for 4 weeks in acupuncture, hypothalamic leptin of model group(0.33±0.02)ng/mg was lower significantly than normal control group(0.47±0.04)ng/mg (P<0.01),hypothalamic leptin of body acupuncture group and electroacupuncture group(0.41±0.03,0.37±0.02)ng/mg were higher obviously than model group(P<0.01 or P<0.05).The body acupuncture group was superior to the electroacupuncture group with significance(P<0.01).7 Effect of different acupuncture methods on serum NPY and hypothalamic NPY of obese rats7.1 Effect of different acupuncture methods on serum NPY of obese ratsAfter acupuncture intervention for 4 weeks,serum NPY of model group(58.10±3.32)ng/ml was higher significantly than normal control group(45.22±2.05)ng/ml (P<0.01),serum NPY of body acupuncture group and electroacupuncture group(49.47±4.76,53.72±4.75)ng/ml were lower obviously than model group(P<0.01 or P<0.05).The body acupuncture group was superior to the electroacupuncture group with significance(P<0.05). 7.2 Effect of different acupuncture methods on hypothalamic NPY of obese ratsAfter acupuncture intervention for 4 weeks, hypothalamic NPY of model group(1.94±0.20)ng/mg was higher significantly than normal control group(1.37±0.17)ng/mg (P<0.01),hypothalamic NPY of body acupuncture group and electroacupuncture group(1.46±0.24,1.68±0.25)ng/mg were lower obviously than model group(P<0.01 or P<0.05).The body acupuncture group was superior to the electroacupuncture group with significance(P<0.05).8 Effect of different acupuncture methods on diameter,area,volume of obese rats' perirenal fat cellAfter intervening for 4 weeks in acupuncture,diameter,area,volume of perirenal fat cell of model group(209.55±19.10,138911.80±26160.94,3.89×106±3.18×105)were bigger than normal control group(115.34±19.29,42825.59±14244.01,6.74×105±2.01×105), the difference has statistics meaning(P<0.01);diameter(173.99±8.84,127.87±14.03)μm,area(95380.77±9665.08,51897.69±11477.08)μm2 , volume(2.70×106±3.53×105,1.50×106±2.64×105)μm3 of perirenal fat cell of electroacupuncture group and body acupuncture group were smaller obviously than model group(P<0.01), the body acupuncture group was superior to the electroacupuncture group with significance(P<0.01).Conclusion:1 Two kinds of acupuncture all can obviously decrease Lee's exponent,perirenal fat weight,pericardial fat weight and epiploon adipose weight.The result proves that these two kinds of acupuncture methods have an obvious effect on weight loss.2 Two kinds of acupuncture all can decrease the content of TG,TC,LDL-C and increase the content of HDL-C in serum,to improve the obese internal LR,IR,hyperinsulinemia state by regulating unusual fat-islet endocrine axis,then regulate the obese's abnormal lipid metabolism.3 Two kinds of acupuncture all have a positive action to Ins, to improve the obese internal LR state by regulating disorders of fat-islet endocrine axis, regulate the glucose metabolism effectively,lower the blood glucose level,and then improve lipid metabolism to achieve weight loss goals.4 Two kinds of acupuncture all can decrease the content of leptin in serum and increase the content of leptin in hypothalamus,to suppress appetite, increase energy consumption,inhibit fat synthesis,then achieve the goal of losing weight.5 Two kinds of acupuncture all can decrease the content of serum NPY and hypothalamic NPY,to reduce hypothalamic NPY neurons in excessive synthesis and secretion,through inhibiting the expression of hypothalamus NPY mRNA,thereby reducing the peripheral and central NPY levels,then make the energy consumption increase,ingest to reduce, and achieve the purpose of losing weight finally.6 Two kinds of acupuncture all can play a role in weight loss,but the effect on the regulation of obese rats' index are different,reducing fat weight,serum TG, TC, LDL-C and Ins content,the electroacupuncture group is superior to the body acupuncture group;reducing Lee's exponent,serum and hypothalamus NPY content,raising HDL-C content and regulating serum and hypothalamus leptin respect, the acupuncture group is superior to the electroacupuncture group.Simple electroacupuncture or simple body acupuncture's effect on weight loss is different,probably due to their regulating organism centre and function of the endocrine are different,use a single acupuncture to treat effectually,or do the two cooperate with the result of use better? Remain to further study.7 In this experiment,we only research electroacupuncture and body acupuncture with a single frequency of stimulation (high frequency) on the comparative study of effects of weight loss,how about body acupuncture and other electroacupuncture stimulation frequencies (intermediate frequency, low-frequency) compared to the slimming effect of impact?To be further completion and study.
