Objective:To explore the new methods of hou to make animal burned models, observed in rats after severe burns application of Chinese medicine Lai Fu Cheng Qi decoction on intestinal barrier and the generation mechanism, providing a theory basis for the application of Lai Fu decoction of Chinese medicine treatment.Method:1.36 wistar rats were randomly selected 30 as the treatment group, given different treatment, each rat were anesthesiaed by intraperitoneal injection of 10% chloral hydrate (40mg/kg), the back hair removal with 4% sodium sulfide, next day in the awake state, the back placed in water 98℃13s, causing the back 30% TBSAⅢ°burned (8×4.5cm2) (confirmed by biopsy) wound, causing third degree burns were randomly divided into two groups (experimental group and scald burn control group), n=15,experimental group received Laifu decoction of Chinese medicine orally, scald control group received normal saline. The remaining six did not burn, given the normal diet as control.2.72h later sterile cardiac puncture wound and blood was sampled for bacterial endotoxin test; the animals were killed to collect peritoneal fluid was inoculated in LB agar for bacterial culture; sterile separation of the liver, spleen, mesenteric lymph nodes, made of homogenized, diluted inoculated in LB agar, and placing CO2 incubator,37℃to do aerobic bacterial culture,48 h later the calculation of colony number of bacteria per gram of tissue colony forming units (CEU) and its standard deviation; the mesenteric lymph nodes, liver and spleen tissue extracts cultured colonies, with automatic bacterial identification instrument Auto24 (U.S.DADE company) and supporting the NC21 identification plates to identify strains.Paraffin sections of conventional systems take jejunum, HE staining, the pathological changes of intestinal mucosa in the optical microscopy.3.Experimental data with mean±standard deviation (x±s)that the use of SPSS 11.5 statistical software.such as variance missing, then the use of approximate analysis of variance (Tamhane's T2);before and after treatment using paired t test (Paired samples Test).Result: 1.The spirit of rats,diet and bowel conditions such as general Burned rats compared with normal group obviously inactive, unresponsive, lazy move, vertical hair, shortness of breath, poor feeding, etc.,early after injury, the majority of the amount and frequency of defecation decreased;traditional Chinese medicine treatment for mental health improvement activity, defecation frequency and volume increased, the quality will be thin; saline treatment group,no obvious improvement in mental condition and bowel movement.2 plasma endotoxin levelsEndotoxin content:burn control group was significantly higher (6724.9±3941.2)than the normal group(1062.3±813.4)(P<0.01), scald burn treatment group(1126.4±719.7)was significantly lower than the control group (P<0.01), burn treatment group was slightly higher than the normal group,but no statistical significance (P>0.05).3.The amount of intestinal bacterial translocationIn addition to the control group, control group and scald scald treatment group had visceral tissue cultures colony formation, visceral tissue burn bacterial translocation in the treatment group (mesenteric lymph nodes 217±147.2,liver67±103.3,spleen 100±167.3)was significantly lower than the control group(mesenteric lymph nodes 850±427.8,liver 150±122.5,spleen 285±183.5),burn treatment group (P<0.01)was slightly higher than visceral tissue bacterial translocation normal group.4. The histological observation of the intestine,Control group, intestinal villus epithelial loose, off, small intestine luminal expansion of small intestinal gland cell erosion to the submucosa to the muscular invasion, treatment group only intestinal glands were enlarged in the normal group were normal.5.Rats ultrastructural changes of intestinal tissueNormal rats, small intestinal mucosa epithelial cells microvilli, rules and closely aligned cells, mitochondria, and clearly, and clearly ridge endoplasmic reticulum structure normal, intercellular Bridges and grain; Burns control rat small intestinal mucosa epithelial cells ultrastructural changes happened obvious, microvilli arrangement, and have relatively sparse scattered in the fall in shape, the epithelial cells of mitochondrial structure of fuzzy, ridge disappears, the endoplasmic reticulum, part of expansion of endoplasmic reticulum, even a saclike intercellular Spaces and bridge structure are unclear, Burn treatment rats, small intestinal mucosa epithelial cells ultrastructure is obviously controls, microvilli hugged tightly, the basic rule, arrangement, but most mitochondrial structure still see part of endoplasmic reticulum, degree of expansion, the intercellular Bridges and basic structure.Conclusion:In this study the preparation of animal model of severe burns, the early Chinese Lai Fu decoction for stomach treatment methods, examine its impact on intestinal barrier function, found that rats with severe burns early enteral nutrition may protect the intestinal mucosa structural integrity, and the Chinese Lai Fu decoction significantly early after severe burns to protect the intestinal mucosa barrier function.
