Objective To investigate the expression of soluble endoglin(sEng) in the Placenta of preecalmpsia.Methods In the study we recruited 67 women with preeclampsia who delivered in the Department of Obstetrics, Affiliated Hospital of Qingdao University Medical College From July 2008 to January 2009, including 31 women with moderate preeclampsia(MPE group) and 36 women with severe preeclampsia(SPE group).And 39 healthy pregnant women were taken as control group. Enzyme-linked immunosorbent assay (ELISA) was used to measure the serum concentrations of sEng. Semiquantitative RT-PCR and Western blot were used to investigate the expression of sEng mRNA and protein in placenta.Results (1) The serum concentrations of sEng in MPE group[(13.66±2.46)μg/L] and SPE group [(16.39±2.05)μg/L], were significantly higher compared with control group [(9.52±0.69)μg/L(P<0.01)].(2) The expression of sEng mRNA in placenta of MPE group (0.82±0.05) and SPE group(0.93±0.05) were significantly higher than that in control group[(0.63±0.04) (P <0.01)].(3) The expression of sEng protein in placenta of MPE group(0.77±0.03)and SPE group (0.89±0.05) were significantly higher compared with control group[(0.63±0.04) (P<0.01)].(4)In MPE and SPE groups, the serum concentrations of sEng were positively correlated with the levels of sEng mRNA and the protein of sEng in placenta [(r=0.8878, 0.7812;P<0.01),(r=0.7598,0.7811;P<0.01)].Conclusions Increased expression of sEng mRNA and sEng protein in Placenta could increase serum concentrations of sEng,which may be involved in the pathoanatomical process of preeclamsia.
