| ObjectiveDetecting of VIP (Vasoactive Intestinal Peptide) in human knee synovial expression and its relationship association with knee pain.MethodsCollected of knee osteoarthritis synovial tissues with patients, and of the normal synovial membrane as controls.Pre-operative geted the VAS pain score and the X-KL stages of the patients.The specimens were first observed by HE staining, morphological,and immunohistochemical staining of synovial VIP expression. According to VAS score were divided into three groups.7 cases of non-pain group (control group) (0 points),16 patients with mild to moderate pain(1-6 points),severe pain in 11 patients (more than 6 points)。By the time compared the synovial lining layer and synovial sublining differences in VIP expression.Results1.Both synovium groups have expressed VIP. Synovial lining layer and sublining expression were no significant difference (P> 0.05). 2.VIP average integrated optical density was 398.48±189.1 in control synovium. VIP average integral optical density was 876.63±395.61 in mild to moderate pain group.VIP average integral optical density was 421.23±202.19 in moderate to severe pain group. VIP expressed in the synovial significantly different between the control group and the mild to moderate pain group (P<0.05),VIP expressed in severe pain group and the mild to moderate pain group, there was significant difference between the two group (P<0.05).VIP was no significant difference in the expression of severe pain group and the control group (P> 0.05).3.The difference was statistically significant at the KL score subgroups divided; group 2 and group 3 they are sig=0.034<0.05 significant difference; Group 2 and Group 4 they are sig=0.043<0.05 significant difference; group 1 and group 3 they are sig=0.01<0.05 significant difference; the other groups had no significant difference.4.There is no linear correlation between synovium VIP expression and the KL score.VIP expression in synovial no linear correlation with pain scores.Conclusions1.Normal and osteoarthritis of the knee joint synovium was expressed in VIP, and in the synovial lining layer and sublining uniform expression..2.VIP expressed in synovial inflammation of mild to moderate pain group than in the control group and the severe pain group, VIP expression in synovial no linear correlation between pain scores.VIP may be involved in the human knee joint synovial membrane in the role of early induced pain. But can not afford a major role in the later pain.3.No linear correlation between synovium VIP expression and the KL score.when early osteoarthritis of the medium-term, X ray changes and the emergence of the nascent lip-like osteophytes and joint space narrowing,the VIP in synovial membrane was higher than the late osteoarthritis.VIP may be relevantand with the development of osteoarthritis. |
