ObjectiveTo investigate the link between Ghrelin and gastric emptying in the diabetic rats at different stages.Methods66 Wistar rats were randomly divided into four group:2-weeks diabetic group, 2-weeks control group, 8-weeks diabetic group, 8-weeks control group, 20-weeks diabetic group and 20-weeks control group. Murine type 2 diabetes model was induced in rat s by intraperitoneal injection of a low dose of streptozotocin plus long-term high fat high caloric laboratory chow. At 2 weeks, 8 weeks and 20 weeks end, the control and model groups as comparison Gast ric emptying half time (GET1/2) and 50 min residual rate were measured by single photon emission computed tomography (SPECT) before sacrifice. The Plasma levels of Ghrelin was detected by Enzyme-linked immunosorbent test (ELISA), observation of gastric tissue Ghrelin immunoreactive cells.Results1 The gastric half-emptying of 2-week diabetic groupwas significantly faster than 2-week normal group[GET1/2:(22.99±2.53 min) vs.(48.76±22.06min),P <0.01)]. There is not difference in DM-8W group and NC-8W group (P> 0.05)and of 20-weeks diabetic groupwas significantly slower than 20-weeks control group[GET1/2: (84.02±27.54 min)vs. 47.79±13.22,P<0.01)].2 In comparision with control group,plasma Ghrelin levels in DM-2w group were significantly increased and significantly decreased in DM-20w group(P<0.05).There were not difference in DM-8W group and control group (P> 0.05)3 Ghrelin immunoreactive cells in DM-2w group increased compared with NC-2W group (P<0.01). There is not difference in DM-8W group and NC-8W group (P>0.05). There is significantly decreased in DM-20W vs. NC-20W (P<0.05). There is no significant difference in control groups(P> 0.05).4 There was negative correlation between plasma Ghrelin levels and gastric half-emptying time.Conclusion1 The changes of gastric motility have varies forms in different stages of diabetesin.The performance in the early accelerates gastric motility.With the progression of diabetes, delayed gastric emptying gradually slows down.2 With the changes in the course of gastric motility, the plasma ghrelin showed a similar trending in gastric motility.3 There was negative correlation between plasma Ghrelin levels and gastric half-emptying time.
