| Objective To investigate association among gastric emptying with ghrelin,obestatin and GHSR,GPR-39 in hypothalamus of diabetic rats.Methods Sixty Wistar rats were randomly divided into three groups:a normal control group(NC group,n=20),a diabetes mellitus group (DM group,n=20),an induced by intraperitoneal injection of streptozotocin(STZ),and an insulin treated group(INS group,n=20). After two and six weeks of STZ injection,gastric emptying was measured by intragastric administration of phenol red; ghrelin and obestatin in the hypothalamus were measured by Enzyme Immunoassay,GHSRmRNA and GPR-39mRNA were measured by RT-PCR.Results After two weeks of STZ injection, gastric emptying(73.63%±8.05%;39.78%±5.07%),ghrelin level (52.31±9.27; 51.13±7.29)and ratio of ghrelin/obestatin (3.77±0.95;2.84±1.04) of hypothesis in DM group and INS group increased significantly compared to those in NC group (32.02%±6.80%; 38.71±11.34; 2.13±0.79,P<0.05). Obestatin level(14.21±2.01) of the hypothesis in DM group decreased significantly compared to those in NC group (21.66±4.74), but GHSR/β-actin (1.26±0.46:0.77±0.21) increased significantly (P<0.05). Gastric emptying was positive correlation with ghrelin level,ghrelin/obestatin and GHSR/β-actin of hypothesis (r=0.49; r=0.63; r=0.73; P< 0.01), but was negtive correlation with obestatin of hypothesis (r=-0.74, P<0.01). After six weeks of STZ injection, gastric emptying in DM group and INS group (78.97%±8.13%; 44.06%±5.06%) increased significantly compared to those in NC group (35.06%±3.91%, P<0.01). Gastric emptying was positive correlation with ghrelin/obestatin of hypothalamus(r=0.40, P<0.05). There was no GPR-39 detected in hypothalamus.Conclusion The rapid gastric emptying may due to the increasing ghrelin and GHSR of hypothalamus in the early status of hyperglycemia,while in the duration of hyperglycemia,to the increasing ratio of ghrelin/obestatin.
