Studies on preparation technology and quality standard and activities of total Flavonoids in Scorzonera Austriaca WildScorzonera austriaca Wild, perennial herbage of Compositae, is widely distributed in the northeast and northwest region of China at an attitude of 1800-3100, such as Shaanxi, Gansu, Ningxia, Qinghai and so on. Scorzonera as a traditional Tibetan medicine (Tibetan Name:DK Badola), whole plant medicine, can be used to anti-inflammatory and set broken bone, dental caries inflammation; topical treatment of wound, ulcer, mastitis embolism. It has been used in Chinese folk medicine for curing hepatitis, and has good effects.This article has made full pharmaceutical research of total flavonoids extracted from Scorzonera austriaca Wild, including several parts as follows:1. Referring to the related literature, we reviewed the latest research progress of Scorzonera austriaca Wild in several aspects such as plant morphology, chemical constituents, pharmacological activity of Scorzonera austriaca Wild and flavonoids.2. Scorzonera austriaca Wild was extracted with 70% ethanol at room temperature, filtrated, and then was removed color impurities by ethyl ether.The extract was subjected to D101 macroporous resin, washed with water to colorless,and 60% ethanol was part of the collection. Dry ethanol in low pression and dry water to abtain Scorzonera austriaca flavonoids.3. Laboratory-made Scorzonera austriaca flavonoids B, purified by semi-preparativehigh performance liquid chromatography,98.9% purity,is used as index components,.we used it to analysis the qualitative and quantitative of total flavonoids in Scorzonera.4. We chose several factors that may influence results of the extraction, including concentration of ethanol, volume, time and times of extraction. First, we did single factor experiments to determinate the extent of these factors, and then orthogonal test showed that the best technology is reflux extration with 60% ethanol(14 folds) for 3 times, half an hour once. Validation experiments were also done to make sure the technology stable and applicable. 5. Macroporous adsorption resin were used to purify the total flavonoids. We referred to the literature and analyzed the structures of those flavonoids, then chose two kinds of resins, D101 and AB-8. By both dynamic adsorption, desorption and adsorption, desorption comparison, therefore, AB-8 was chosen. Purification of the impact of various factors:the sample volume, flow rate, adsorption time and cleaning water were investigated, the result was as follows:extracted in accordance with the best extraction process of total flavonoids Scorzonera austriaca extract to 1.0BV/h flow rate through the pretreatment, AB-8 macroporous resin column, adsorption 6h, with 10BV of distilled water to 2.0BV/h flow rate wash after elution with 60% ethanol, flow rate 1.0BV/h, collecting eluate, evaporated to dry, that Scorzonera total flavonoids were extracted.6. In accordance with the Pharmacopoeia of the content, from the identification, examination and determination of the three aspects of stab ginseng saponin extract the required quality standards to do:after the Scorzonera austriaca purified flavonoid B as the reference standard, the establishment of a TLC method; determined Scorzonera austriaca total flavonoids extracted from the water, total ash, acid insoluble ash, and develop limits; to Scorzonera austriaca flavonoid B for the index components, using high performance liquid chromatography (HPLC) to establish the total flavonoids Scorzonera austriaca. Determination to provide for total flavonoids in Scorzonera austriaca flavonoids content of not less than B 29.24%.7. According to the depth study of the activity of total flavonoids extracted from the Scorzonera austriaca. Experiment is divided into six groups:normal group, carbon tetrachloride model group, positive medicine group (DDB), low dose group (80mg/kg), middle dose group (160mg/kg) and high dose group (320mg/kg), the experimental data with group T test results show that total flavonoids of Scorzonera austriaca have hepatoprotective activity.
