Scorzonera austriaca Willd,a perennial herb belonging to the genus Artemisia,is a traditional Tibetan medicine.In China,it is mainly distributed in the northeast,north China and parts of the northwest,such as Jilin,Shanxi and Ningxia.Used for the treatment of sores,mastitis,etc.The related pharmacological experiments showed that the flavonoids of Scorzonera austriaca Willd had hepatoprotective activity,and the new compound flavonol-caffeoyl derivatives were likely to be the basis for the liver protection of Scorzonera austriaca Willd.The separation and enrichment are of great necessity.In this experiment,the flavonol derivative was used as the raw material to determine the contents of the new flavonoids,and the macroporous resin was used to separate and enrich the flavonyl caffeoyl derivatives,and the process parameters were determined.This improved the relevant content of quality control of the Scorzonera austriaca Willd,and also provided a certain foundation for the study of the anti-hepatitis virus pharmacological activity.The research results are as follows:1.Extraction and separation the flavonoids from Scorzonera austriaca Willd.Take 15kg dry Scorzonera austriaca Willd,70%ethanol leaching,ethanol recovery of the extract,combined water layer,adsorbed by D101 macroporous resin column,eluted with 60%ethanol and collected,vacuum recovery of ethanol,drying,Scorzonera austriaca Willd crude flavonoid extract 325g.By means of silica gel column chromatography,ODS column chromatography and other separation and purification methods,two flavonoids caffeoyl derivatives were obtained:Luteolin 8-C-(6"-O-caffeoyl-?-D glucopyranoside),Apigenin 8-C-(6"-O-trans-caffeoyl-?-D-glucopyranoside).2.To establish a method for determining the content of flavonolyl caffeoyl derivatives.Luteolin 8-C-(6"-O-caffeoyl-?-D glucopyranoside)was used as the reference substance.The isocratic concentration of the reference substance was analyzed by HPLC.The detection wavelength was 337 nm and the peak area was recorded.The regression equation was used to determine the content of caffeoyl derivatives in total flavonoids.3.Separation and enrichment caffeine derivatives of Flavonoids from Scorzonera austriaca Willd.Through dynamic separation,static test,adsorption and analytical performance,it was found that Hp20-type macroporous resin has a better enrichment effect,so the Hp20 was selected.4.Determination of elution process parameters of Hp20 macroporous resin.Results:When the sample solution flow rate is the same,the relatively high concentration of total flavonoid solution of leek on the adsorption rate is faster,the quality of the blank resin and the loading resin is not less than 2:1,first remove with 30%ethanol non-target compounds were eluted,then with a 50%ethanol eluent at a rate of 2 BV·h-1 and the target compound content exceeds 90%.
