ObjectiveTo observe the change of serum ceramide in type 2 diabetic rats and its relationship with oxidative stress.MethodsThe male Sprague-Dawley (SD) rates were randomly assigned into normal group (NC) rats(n=15) and model group (DM) rats(n=45). Normal group rats were fed with normal diet, while model group rats were fed with a high-sucrose-high-fat diet and after 6 weeks a low dose of streptozotocin (STZ,30 mg/kg) was injected to induce type 2 diabetic model. And then the model group rats were divided randomly into two groups again:DM untreatment group(DMU) and DM treatment group(DMT). DM treatment group(DMT) rats were induced by abdominal injecting myriocin 0.3mg/kg Qod, while normal group and DM untreatment group(DMU) were induced by abdominal injecting 0.9%NaCl. The nomal group rats were fed with the normal diet, DM untreatment group(DMU) rats were fed with a high-sucrose-high-fat diet continue.All rats were executed after intervene 12 weeks, and collected the fasting blood before all rats were executed. The HbAlc, insulin,ceramide,SOD,GSH-Px,levels of lipid and hepatic,renal functions were measured by the blood. Results1 The SD rats were successed induced into the type 2 diabetic model by fed with a high-sucrose-high-fat diet and a low dose of STZ (30mg/Kg) was injected. The FBS of all the type 2 diabetic model rats were≥7.0mmol/L, the PBS were≥11.1mmol/L. Compared with the normal group, insulin resisitance index in DMU and DMT were significantly higher (p<0.05),and the DMU were much higher than the DMT (P<0.05)2 Compared with normal group, FBS and PBS were significantly higher in DMU and DMT before execution (p<0.05), while the DMU were much higher than the DMT (P<0.05)3 Compared with normal group, HbAlc were significantly higher in DMU and DMT(p<0.05), while the DMU were much higher than the DMT (P<0.05). The injury of liver in both DMU and DMT were higher than normal group (p<0.05), while the DMU were much higher than the DMT. The injury of renal in both DMU and DMT were higher than normal group (p<0.05),but there were no difference between the DMU and the DMT (P>0.05). The levels of lipid in both DMU and DMT were higher than normal group (p<0.05), and the levels in DMU were higher than the DMT (p<0.05)4 Compared with normal group, the levels of serum ceramide were significantly higher in DMU and DMT (p<0.05), and the levels in DMU were higher (p<0.05)5 Compared with normal group, the levels of serum SOD and GSH-Px were significantly lower in DMU and DMT (p<0.05), otherwise the levels in DMU were much lower (p<0.05)6 As the correlation analysis showed, ceramide and SOD were significantly linear correlated (p<0.05), the Pearson coefficient correlation was-0.649. While, the same linear correlation was exsisted between ceramide and GSH-Px, the Pearson coefficient correlation was-0.705.Conclusion:1 The levels of serum ceramide in type 2 diabetic rats were higher.2 The oxidative stresses strengthen in type 2 diabetes rats, and it can be weakened by myriocin in the way of blocking the synthesis of serum ceramide in type 2 diabetic rats.3 The study above showed that ceramide may participate in the oxidative stress process in type 2 diabetic rats.
