OBJECTIVE To establish a model of obese rats with type 2 diabetes by feeding a high-fat,high-sugar diet combined with a small dose of streptozotocin,and to randomly divide the model into experimental and control groups.The experimental group was given Saxagliptin intervention.Finally,by detecting the change of serum ceramide concentration in rats,we observed the changes of ceramide concentration in each group after intervention with saxagliptin,and explored the effect of saxagliptin on oxidative stress in type 2 diabetic rats.METHOD Sixty male Sprague Dawley rats were randomly divided into the normal control group and the model group.Streptozotocin was divided into diabetic control group and Shah Glenn Dean treatment model of type 2 diabetic rats induced by special high fat diet.The patients in the treatment group were treated with saxagliptin,and the fasting blood glucose was measured with a blood glucose meter.Then the changes of indexes such as liver function,renal function,blood lipid and fasting insulin were detected by an automatic biochemical analyzer.The serum ceramide,SOD concentrations were detected by Elisa examination.RESULT(1)The indexes of blood glucose,blood lipids,and insulin resistance were significantly higher in the DM group and the DM-sax group than in the NC group.The above indexes in group DM were all lower than that in group DM,and the difference was statistically significant.(2)compared with that in group NC,the weight of the DM group and the DM-sax group decreased;the weight changes of the DM-sax group before and after the monitoring test were not statistically significant.(3)The levels of ceramide in serum of rats in DM group and group were significantly higher than those in normal group.The serum ceramide level in DM group was lower than that in DM group,The difference was statistically significant.CONCLUSION With the development of animal models of type 2 diabetes with obesity,serum cholesterol,LDL,and ceramide concentrations increased,and SOD levels decreased.After intervention with saxagliptin,the above indicators showed a downward trend,and SOD levels increased,indicating that Shagley was Ting may reduce lipotoxicity and reduce oxidative stress in the treatment of obese patients with type 2 diabetes.
