Objective Through the Investigation of DYS576 and DYS64 loci of genetic polymorphism in the Han population of Shanxi, study their gene polymorphism and population distribution in Shanxi Han male population, in order to provide reference data for forensic individual identification, paternity testing and to explore its forensic application.Methods We randomly selected 205 unrelated healthy male individuals'blood samples 1ml of Chinese Han population of Shanxi. One man's blood and several kinds of his organization tissues were selected, too, for the conduct of identity test. We randomly selected 30 unrelated healthy individual women's blood sample 1ml, EDTA anticoagulant, used for male-specific tests. Thirty cases of two generations of parent-child whose relationship has been confirmed were collected, their blood samples were used for mutation observe. We collected several common animal's tissue or blood samples used for species test. The genomic DNA from the material above were extracted, and amplified by PCR with the loci DYS576 and DYS641 primers. The products of PCR were analyzed with polyacrylamide gel electrophoresis. Each allele of these Y-STR loci was sequenced and the allele ladders were constructed. Alleles of these Y-STR loci were nominated according to recommendations of the International Society of Forensic Genetics. Finally the data was statistical analysed, come out the polymorphism distribution of these two loci in Han population of Shanxi.Results Two hundred and five cases in Shanxi Han male of DYS576 locus has been detected seven alleles, named DYS576*15, DYS576*16, DYS576*17, DYS576*18, DYS576*19, DYS576*20 and DYS576*21, the gene diversity are 0.0488,0.1024,0.2780,0.2439,0.2146, 0.0976 and 0.0146 respectively, their personal identification capabilities (DP) and non-parent excretion rate (PE) are 0.7984; DYS641 locus has been detected six alleles, named DYS641*6, DYS641*7, DYS641*8, DYS641*9, DYS641*10 and DYS641*11, the genetic diversity are 0.0927,0.4780,0.0634,0.1024,0.2098 and 0.0537 respectively, their personal identification capabilities (DP) and non-parent excretion rate (PE) are 0.7050. The haplotypes composed of these two loci has found 28 species in 205 Chinese Han male individuals of Shanxi Province, its genetic variability is 0.9423, its personal identification capabilities (DP) and non-parent excretion rate (PE) are 0.9423. The blood and different tissue test results from one healthy body with two loci were consistent typing. Tirty cases of women, no amplification products of DNA sample was found. Tirty cases observed no mutation in two generations of family. Detection of male animals, no amplification products was found.Conclusion DYS576 and DYS641 loci have a high level of genetic polymorphism and can be used in forensic science and human genetics research.
