Objective: Quantify the intensity of noxious stimulation by neural electrophysiology, access to analysis skin,muscle and fascia's pain threshold and sensitivity of rat model of tension headache precisely. To detect the plasma NO/NOS levels of patients with tension-type headache and rat model of tension headache, access to research the relationship between NO/NOS and TTH.To study the Roujin Decoction's influence on the plasma NO/NOS levels of rat model of tension headache, access to get the exact therapeutic mechanism of action.Methods: Animal experiments: Modeling method is both semispinal neck intramuscular injections of ATP. Observed amplitude and latency of rats'JOR before and after administration. After the test,blood from abdominal vein of all rats to detect the plasma NO/NOS levels.Made 10% brain homogenate from the left brain to detect the NO/NOS levels.clinical trials 1: Select 41 patients with CTTH, blood to detect the plasma NO/NOS levels and compare them of healthy's plasma NO/NOS levels.clinical trials 2: Divided 41 patients with CTTH in clinical trials 1 randomly into two groups:21 patients were treated with Roujin Decoction as the treatment group, 20 patients were treated with fufangyangjiaokeli as the control. Via analyze the change of patients'headache index to research the clinical effects of Roujin Decoction after 28 days.Results: (1) The plasma and brain homogenate NO/NOS levels of rat model of tension headache increase abnormally(compared with the control group,p<0.01). Roujin Decoction and fufangyangjiaokeli can reduce plasma and brain homogenate NO/NOS content( compared with model group respectively,p<0.05). There are not statistically significant differences in the plasma NO/NOS content between patients with CTTH and the healthy(P>0.05).(2) Latency of rats'JOR changed significantly in NOS inhibitor group before and after ATP injection(P<0.05). And proved that NOS inhibitor can reduce the skin,muscle and fascia's sensitivity of rat model of tension headache.(3) Pain threshold of rats after ATP injection in model group , compared with which before ATP injection, tend to reduce((P<0.05). Rats after ATP injection in model group have a lower pain threshold(compared with the control group,p<0.05).(4) Roujin Decoction can extend rat model's incubation period of pain, reduce the skin,muscle and fascia's sensitivity(compared with model group ,p<0.05).Conclusion: (1)The mechanism of tension headache is influenced by the drop of skin,muscle and fascia's sensitivity .(2)NO/NOS are closely related with the incidence of tension headache. NO/NOS have impact on skin and fascia's sensitivity ,through which they participat tension headache.(3)Roujin decoction can effectively relieve headaches, improve clinical symptoms.(4) Roujin decoction can accommodate the plasma and brain homogenate NO/NOS levels.And this process is one part of its action to tension headache.
