Objective:To establish a model of chronic abacterial prostatitis,CAP of SD-rat and to investigate difference of intracellular calcium ion concentration of smooth muscle between model group and control group;to evaluate the role of the intra- cellular calcium ion concentration in the model group.Method: An antigen solution was made by mixing the SD rats, purified prostate protein and the Freund's complete adjuvant(FCA).The model of CAP was established by injecting the solution subcutaneous with a multi-point way. Every rats of the CAP group was injected 0.5ml DPT througt intraperitoneal.Dissected prostate in aseptic condition after 8 weeks. Select the back lateral lobe in the clean bench and stripping the prostate capsule, blood vessels, fat and other tissues.Part of tissue used for HE staining, The bigger part is for cell culture. PSMCs were cultured in vitro and purified. Immunocytochemistry staining were used to identify the expression of specific smooth muscle antibody. The cells were passed into the laser confocal scanning microscope (LSCM) special dish on the second generation.LSCM was used to dynamic scan the cells which were incubated with FLUO-3AM for 30 minutes inside the incubator. Observed the changes in the fluorescence intensity of the cells. Useing the Leica image analysis system to analyze the results.Result: A large number of spindle-shaped and polygonal cells were cultured in vitro,the cells grew as a whirlpool when became confluent,accorded with the growth characteristic of smooth muscle,and immunocytochemistry staining result confirmed the PSMCs with positive expression ofα-SMA.In CAP group,prostate tissue was infiltrated by lymphocytes and plasma cells , consistent with pathological characteristics of chronic inflammation. The control group without prostatitis.The difference of the intensity of fluorescence between the model group and control group scanned by LSCM was observed.The intensity of fluorescence was80.39±9.00and 27.95±10.04, respectively.The difference has statistical significance(p<0.01).Conclusion:The intracellular calcium ion concentration of PSMCs increases in CAP of SD-rat.The increased calcium ion in PSMCs of CAP may enhance the constriction of PSMCs, which subsequently increase the sensibility of pain and causes the symptom, e.g. low abdominal pain.
