| Objective:The diagnosis of acute pancreatitis(AP)is usually confirmed by a significant increase of the serum amylase and/or lipase level,and acute pancreatitis is often associated with impaired cardiovascular function.This depressed contractility has been attributed to altered[Ca2+]i.The objective of present study was to examine effects ofα-Amylase on ICa-Lin ventricular myocytes isolated from adult rat and single smooth muscle cells isolated from coronary arteries to figure out the possible reasons of the depressed heart function during the AP.methods:Whole-cell Patch clamp technique was used in this study.Results:α-Amylase can significantly increase ICa-Lin ventricular myocytes isolated from adult rat and single smooth muscle cells isolated from coronary arteries respectively in a dose-dependant manner.Conclusion:Our data indicate that(1)α-Amylase can significantly increase ICa-Lin ventrieular myocytes isolated from adult in a dose-dependant manner which could be the possible reason of ca overload of myocytes during the process of AP and contribute to cardiac failure;(2)α-Amylase can significantly increase ICa-Lin single smooth muscle cells isolated from coronary arteries from rat in a dose-dependant manner which could increase the vascular resistance then reduce the blood flow in the heart and and contribute to cardiac dysfunction during the process of AP. |
