According to the basic pathogenesis of accumulated dampness-heat syndrome for long,to cause the deficiency transformed from excess, blood-stasis resulted from deficiency of refractory nephropathy syndrome. The famous doctor of academic institution of TCM in Sichuan province developed a related formula-LiShen capsule which has efficacy of clearing away heat and promoting diuresis, and detoxification, consolidates the constitution, which is used for treating RNS.Objective:To study on pharmacological action of LiShen capsule which is used for RNS with the model of whole animal experiment, it aimed to explore and further clarify its mechanism of action, and look forward to provide scientific basis for clinical reasonable using of medicine.Methods:This subject mainly observed the pharmacological action of LiShen capsule treating RNS by several aspects as following:(1)To observe the therapeutic effect of LiShen capsule on rat model of nephritic syndrome induced by adriamycin, We developed the nephropathy rat model by unilateral nephrectomy combined repeated injecting ADR, and examined the 24 hours urinary protein, serum biochemical, hemorheology, the renal tissue's pathological changes. (2)Diuretic action experiment, according to the water loading rat model to observe the diuretic action of the drug, and assay the electrolyte in the urine. (3) Immunoregulation effect, we developed the mice immunosuppression model inducing by the cyclophosphamidum, to observe the immunoregulation effect of the drug on the macrophage phagocytic function test and hemolysin test. (4) Promote blood circulation to remove blood stasis experiment, according to the experiments of the clotting time of mice and coagulation function of rat to observe the promotion of blood circulation of the drug. (5) Antipyretic effect experiment, according to the rabbit fever model inducing by the LPS to observe the Antipyretic effect of the drug.Results:(1)In anti-nephropathy test, compared with the model group,LiShen capsule high dose group can reduce the 24 hours urinary protein steadily and persistently (P<0.01). the high and middle group can also increase the content of serum albumin, reduce the TC&LDL and CR(P <0.05). Each group can reduce the low-muting blood viscosity in various degree(P<0.05, P<0.01). The high dose group can also notablely reduce the RBC aggregation index(P<0.05). Under light microscope, the high dose can improve the pathological changes of glomeruli& tubulointerstitium. (2)In the Diuretic action test, compared with the blank control group, the high dose group has the obivious diuretic action(P <0.05, P<0.01). Each group can increase the excretion of k+ na+ cl+ in the urine(P<0.05, P<0.01). (3) In the immunoregulation effect test, compared with model group, Each group can notably improve the macrophage phagocytic function (P<0.05, P<0.01), however, has no effect on the generation of hemolysin. and can also increase the thymus&spleen index of the mice immunosuppression model. (4) In the promote blood circulation to remove blood stasis test, compared with the blank control group, Each group can lengthen the mice clotting time and ameliorate the rat coagulation function in various degree (P<0.05, P<0.01). (5) In the antipyretic effect test, Each group has the lowerâ–³T andâ–³Tmax than the model group(P<0.05, P<0.01).Conclusion:LiShen capsule has the clear effect on the RNS in animal experiment. It can reduce the urinary protein and cholesterin, elevate the hypoproteinemia,protect the renal function, reduce the pathological damage. and also has certain effect on clearing away heat and promoting diuresis, tonifying the kidney, removing blood stasis and regulating immune function.
