Objective:Chrysanthemum indicum is the dry capitulum of Chrysanthemum indicum L. It contained in the "Appendant records of famous physicians", stated "bitter, acrid, slightly cold", with the effect of "detoxification and calming the liver". Modern pharmacological studies have shown that it had the effect of broad-spectrum anti-bacterial and anti-inflammatory, antiviral, lowering blood pressure, antioxidant and other pharmacological effects.In order to develop the Chrysanthemum indicum resources, evaluate the quality of it better. This paper investigated the influcence of extraction and purification to Chrysanthemum indicum extract components, the stability of linarin under different conditions; established IR spectra and HPLC fingerprint by different content of linarin in Chrysanthemum indicum; and research antioxidant effect of water extraction, with Chrysanthemum indicum which has different content of linarin.Methods and results:1. Study on the impact of extraction to the components in Chrysanthemum indicum extractionUsed of four factors and three level orthogonal test to study the extraction process, with impact of the total flavonoids and linarin in Chrysanthemum indicum extraction. The optimized process parameters were as follows:50% ethanol,80℃, for 3 times,1 hour each time. 2. Study on the impact of purifying to the components in Chrysanthemum indicum extractionStudy on the impact of total flavonoids recovery in Chrysanthemum indicum extraction, by different adsorption and desorption conditions of DM130 resin. The purification condition was as follow:The concentration of total flavonoids of Chrysanthemum indicum was 2.78 mg/mL, pH3-4, the adsorption velocity was 2 mL/min. After 30 min, washed with water first, then with 80% alcohol being 5 times as much as the resin volume, and the elution velocity was 2 mL/min. The recovery of total flavonoids was 79.19%, and the purification rate was 37.31%.3. Study on the stability of linarin in Chrysanthemum indicumTo determine the content of linarin in Chrysanthemum indicum, by different processing procedure, different storage conditions and time, inspect the influence of pH value and additives to the content of linarin in Chrysanthemum indicum solution, as well as drying temperature and drying methods to the content of linarin in its extracts. The results showed that the linarin had greater losses in carbon products, had little effect on different storage conditions and time. The pH and additives had differnet influences on the content of linarin in Chrysanthemum indicum solution. The content of Linarin in extracts would loss increased when the drying temperature increment and time prolongation, but it could be preserved better by spray-drying.4. Study on the IR spectra of Chrysanthemum indicum of different content of linarinSeven identified characteristic peaks of Chrysanthemum indicum were marked:2924 cm-1,1737 cm-1,1655 cm-1,1514 cm-1,1376 cm-1,1245 cm-1,1051cm-1. The Chrysanthemum indicum of not detected linarin and bought from Shuanghe, Hongshan, Hubei, both had absorbed in 414cm-1,1414cm-1,1641cm-1, but the Chrysanthemum indicum from other sources had no absorption in the three waves. The similarity between different content of linarin in Chrysanthemum indicum is different. The second derivative spectra can distinguish the similar IR spectrum better.5. Study on the HPLC fingerprint and use in the preparation processEstablished the Chrysanthemum indicum fingerprint by RP-HPLC, and researched the fingerprint of the Chrysanthemum indicumwith different content of linarin, then evaluated the similarity. The results showed that the HPLC draft had great difference between them.The chromatographic condition was used to inspect the processing and drying process. The results showed that the steam products and liquor products HPLC draft had good similarity with raw materials, but the similarity of fried products was relatively poor; it could preserve chemical composition of Chrysanthemum indicum well by spray drying method.6. Study on the antioxidant in water extract of Chrysanthemum indicumUsed SOD activity and MDA content as pharmacological targets, test strictly in accordance with the detection kit, to study the antioxidant effect in water extract of Chrysanthemum indicum with different content of linarin. The results showed that they had a certain antioxidant effect, but there had no significant difference between the water extract with different content of linarin, the high content of linarin had a better effect.Conclusion:The results showed that the content of linarin in Chrysanthemum indicum preparations can be affected by extraction purification additives,pH value,drying temperature and drying time. There is a certain difference in IR spectrum and HPLC fingerprint of Chrysanthemum indicum with different content of linarin. The results could provide reference for improving the content of linarin in Chrysanthemum indicum preparations, evaluating quality and developing resources of Chrysanthemum indicum.
