Chrysanthemum indicum L belongs to the family Compositae. The ethanolic extract, partitioned fractions by different polar solvents and purified parts by macroporous resin were prepared and tested through a series of antioxidant experiments from Chrysanthemum indicum L. We hope to find higher antioxidant fraction and get its components through suitable separation techniques. Then these components will be tested and compared for antioxidant capacity. Finally, extracts and parts purified by macroporous resin were analyzed to get a preliminary antioxidant material basis of wild chrysanthemum. This thesis is divided into four parts, and the specific contents are as follows:1. Antioxidant capacity test of extracts from Chrysanthemum indicum LChrysanthemum indicum L was extracted 3 times, each time 2h by 80% ethanol with 8 times the amount of meterial. This ethanol extract was further patitioned by petroleum ether, ethyl acetate, n-butyl alcohol sequently. Then 4 kinds of antioxidant methods were applied including anti-DPPH and anti-superoxide radical scavenging test, determination of Fe ions reducing power and lipid peroxidation of linoleic acid antioxidant system test. The results show that n-butanol layer, the ethyl acetate layer and purified product by macroporous resin have relatively better antioxidant capacity.2. Separation of single components in antioxidant parts from Chrysanthemum indicum LDifferent chromatographic methods using positive silicagel, C18 reverse silicagel and Sephadex LH-20 dextrangel and so on, were applied to isolate the single components in antioxidant parts. Through HPLC, LC-MS, NMR analysis, 6 compounds were identified as Linarin, luteolin, caffeic acid, 1,3-caffeoylquinic acid, 3,4-caffeoylquinic acid, 3,5-caffeoylquinic acid.3. Antioxidant capacity test of single components form Chrysanthemum indicum LThe isolated compounds were tested and evaluated for antioxidant activity. The results showed that single compounds have significant antioxidant capacity, including 1,3-caffeoylquinic acid, 3,4-caffeoylquinic acid, 3,5-caffeoylquinic acid, caffeic acid, chlorogenic acid, luteolin.4. Preliminary analysis of antioxidant components in Chrysanthemum indicum LFor preliminary analysis of antioxidant material basis of Chrysanthemum indicum L,we analyse the different extracts and purified product of macroporous resin by HPLC. Analysis showed that the parts which have better antioxidant activity such as n-butyl alcohol or ethyl acetate contain higher 1,3-caffeoylquinic acid, 3,4-caffeoylquinic acid, 3,5-caffeoylquinic acid, luteolin. This result is agreed with that of activity test. |
