Objective1. Analysis blood sample results of glucose 6-phosphate isomerase (GPI) antigen concentration in patients with rheumatoid arthritis (RA), patients with other causes induced arthritis and healthy people, then analysis the blood results of anti-cyclic citrullinated peptide antibodies (anti-CCP antibodies), rheumatoid factor (RF), C-reactive protein (CRP), erythrocyte sedimentation rate (ESR) in patients with rheumatoid arthritis (RA), patients with other causes induced arthritis and healthy people, to explore the GPI antigen diagnostic significance in patients with RA.2. Analysis the correlation for the GPI, CRP, RSR and RF with the RA patients, to explore the correlation between these test items.3. Analysis the GPI antigen concentration in the each type of traditional Chinese medicine syndrome of RA, to summarize the relationship between GPI antigen concentration levels and traditional Chinese medicine syndrome of RA.Methods1. Collected blood samples of that 21 patients with RA,34 cases of other reasons induced arthritis (including 23 cases of osteoarthritis,11 cases of gouty arthritis) and 12 cases of healthy people controls, then detected the quantification of the concentration of the GPI antigen in serum by the double antibody enzyme linked immunosorbent assay, qualitative detected the anti-CCP antibodies in serum by the indirect enzyme linked immunosorbent method, detected the RF and the CRP in serum by the immunoturbidimetric method, detected the ESR in blood anticoagulant by the optical transmission method, and the results were statistically analyzed.2. Analysis of the test items on the sensitivity and specificity of RA diagnosis, and calculate the test items the positive predictive value and negative predictive value on the RA group, and do statistical Analysis for the positive rate of the test items in the RA patients.3. Analysis of the project correlation between GPI, anti-CCP, CRP test items and RF test item, and do statistical analysis.4. To approach the relationship of TCM syndrome typies of RA with levels of GPI antigen concentration.Results1. The RA patient group GPI concentration was (1.67±1.56) mg/L, mean confidence interval was (1.38,1.94) mg/L;median was 0.91 mg/L, the median confidence interval was (0.78,1.51) mg/L; lower quartile P25 was 0.43 mg/L, upper quartile P75 was 2.92 mg/L, quartile range was 2.49 mg/L. The median concentration of RF was 65.50 IU/mL, lower quartile P25 was 27.10 IU/mL, upper quartile P75 was 80.47 IU/mL, quartile range was 53.37 IU/mL. The median of CRP was 14.90 mg/L, lower quartile P25 was 5.15 mg/L, upper quartile P75 to 60.70 mg/L, quartile range was 55.55 mg/L. The mean of ESR was (61.66±39.90) mm/h, median was 75.00 mm/h, lower quartile P25 was 22.00 mm/h, upper quartile P75 was 99.50 mm/h, quartile range was 77.50 mm/h. Other causes induced arthritis patients, the median serum concentration of GPI was 0.01 g/L, lower quartile P25 was 0.01 mg/L, the median confidence interval was (0.01,0.01) mg/L, upper quartile P75 is 0.08 mg/L, quartile range was 0.07 mg/L. The median concentration of RF was 17.00 IU/mL, lower quartile P25 was 6.00 IU/mL, upper quartile P75 was 25.00 IU/mL, quartile range was 19.00 IU/mL. The median CRP was 3.00 mg/L, lower quartile P25 was 3.00 mg/L, upper quartile P75 was 9.50 mg/L, quartile range was 6.50 mg/L. The median ESR was 23.00 mm/h, lower quartile P25 was 13.50 mm/h, upper quartile P7b was 34.00 mm/h, quartile range was 20.50 mm/h. Healthy control group, the median serum concentration of GPI was 0.01 mg/L, the median confidence interval was (0.01,0.01) mg/L, lower quartile P25 was 0.01 mg/L, upper quartile P75 was 0.02 mg/L, quartile range was 0.01 mg/L. The median concentration of RF was 6.00 IU/mL, lower quartile P25 was 6.00 IU/mL, upper quartile P75 was 12.10 IU/mL, quartile range was 6.10 IU/mL. The median of CRP was 3.00 mg/L, lower quartile P25 was 3.00 mg/L upper quartile P75 was 5.50 mg/L, quartile range was 2.50 mg/L. The median of ESR was 10.00 mm/h, lower quartile P25 was 6.00 mm/h, upper quartile P75 was 17.00 mm/h, quartile range was 11.00 mm/h.2. The positive rate of GPI in the RA group was 80.95%, which was significantly higer than that of the RF in the RA group the 61.90% positive rate (P<0.05), the positive rate of ESR in the RA group was 76.19%, which was significantly higer than that of the positive rate of RF in the RA group (/?<0.05), GPI and ESR positive rate of diagnosis of RA were higer than RF positive rate of 61.90%. The positive rate of anti-CCP in the RA group was 71.43%, which was not significant difference in positive rate with that of the RF in the RA group(P> 0.05). The positive rate of CRP in the RA group was 61.90%, which was not significant difference in positive rate with that of the RF in the RA group (P> 0.05).3. GPI, CRP, ESR and RF did not form a linear relationship between the test datas. In RA patients, the correlation coefficient of GPI and RF was 0.14, the correlation coefficient of CRP and RF was 0.27, the correlation coefficient of ESR and RF was 0.31. When the GPI concentration in the positive critical value was 0.20 mg/L, the corresponding RF values can be in the reference range, also can be up to 187.13 IU/mL. when the GPI concentration was the median 0.91 mg/L, the corresponding RF values was mostly higher than the reference range.4. The wind cold dampness resistance type and depressed type of GPI concentration levels may be relatively higher, the GPI concentration levels of the wind cold dampness resistance type with RA patients were a few higher than the other syndromes (K0.05).Conclusion 1. GPI antigen concentration levels increased in the serum of RA patients, can be used as a new test project in auxiliary diagnostic of RA.2. GPI antigen concentration levels in the wind cold dampness resistance type was higher than the other types, GPI may be as the auxiliary test item to judge the traditional Chinese medicine syndrome type of RA.
