[Objective]:To study the expression of f HIF-2a and VEGFin colorectal carcinoma and the adjacent tissues, as well as their mutual relationships, then to investigate their roles and probable mechanisms in the pathogenesis of colorectal carcinoma.[Methods]:The immunohistochemical SP technique was applied to detect the expression and distribution of HIF-2a and VEGF as well as microvessel density (MVD) in 40 cases of colorectal carcinoma and their corresponding normal colorectal tissues served as the control group. The results were statistically analyzed.[Results]:The positive expression rates of HIF-2α,VEGF and CD34 in colorectal carcinoma were significantly higher than that in the control group.(P<0.05) The expression of HIF-2a protein was mainly localized in the cytoplasm, as well as in some nucleus.especially on the edge of the tumor and nearby the regions that have a large number of palingenetic vessels. The expression of VEGF protein was mainly localized in the entosarc and cytomembrane, presented a tan or light brown color. The positive expression of CD34 was mainly in the vascular endothelial cells.It also could be seen in the nests and distributed dispersedly.. The expression of HIF-2a was pos—itively correlated with the expression of VEGF and the value of MVD closely(P<0.05).. The positivity rates of HIF-2a and VEGF were closely related with TNM staging and tumor size of colorectal carcinoma repectively. Three ones have nothing to do with the degree of tumour differentiation. [Conclusion]:1.The positive expression rates of HIF-2a and VEGF in colorectal carcinoma are significantly higher than that in the control group.It indicates that the three ones play an important role in the carcinogenesis and aggression of colorectal carcinoma.2. HIF-2a is overexpressed in colorectal carcinoma, and its expression is closely correlated with the expression of VEGF and the value of MVD. It may be involved in the angiogenesis of human colorectal carcinoma by upregulating the expression of VEGF, and play an important role in the carcinogenesis and aggression of colorectal carcinoma.3.. The positivity rates of HIF-2α, VEGF and the value of MVD are closely related with TNM staging and tumor size of human colorectal carcinoma respectively.4. The positive expression rates of HIF-2α,VEGF and CD34 in colorectal carcinoma have nothing to do with the differentiation of colorectal carcinoma.
