[Objective] To explore the effect of resveratrol on serum anti-oxidase activities improvement and anti-atherosclerosis in atherosclerosis and hypercholesterslimia model rabbits.[Methods] Sixty male Japanese white rabbits were randomly divided into 6 groups (10 rabbits for each group):group A (normal control group) fed with normal feedstuff; group B,C (pathological control group) fed with high cholesterol feedstuff; group D (treated with low dosage resveratrol 5mg/kg-d), group E (treated with mid dosage resveratrol 15mg/kg-d) and group F (treated with high dosage resveratrol 45 mg/kg-d) fed high cholesterol feedstuff combined with resveratrol. Only group B was fed for 10 weeks, the rest groups were fed for 20 weeks. From the end of the 10th week, the D,E,F groups were feeding accompanied by oral administration of resveratrol for ten weeks. Serum total cholesterol (TC), triglyceride (TG), low density lipoprotein-cholesterol (LDL-C) and high density lipoprotein-cholesterol (HDL-C) of the rabbits were detected by biochemical method at the first day and the end of the20th week. At the end of the 20th week, animal were killed, the hearts and aorta were taken to do the Hematoxylin-Eosin (HE) staining. The activity of GS,GSH-px,GST,CAT,T-SOD,γ-GCS,GSH,MDA in serum were determined.[Results]1 The atherosclerosis model with rabbits had been successfully established with high fat diet at the end of 10th week 2 The anti-atherosclerosis effect of resveratrol:there is no AS plaque found in the control group. The terrible AS pathological changes can be found in pathological group. After treated with resveratrol, the lesion improved. The effect in group E and F are more significant than group D. After established the animal model, the blood lipid level of pathological group significantly increased compared with control group (P<0.01); after treated with low does resveratrol, the level of TC,TG,LDL-C decreased compared with pathological group (P<0.01), otherwise the level of HDL-C incresed. The effect in group E and F are more significant than group D.3 One possible mechanism of resveratrol in anti-atherosclerosis effect--anti-oxdative stress:the activity of GSH-PX,CAT,T-SOD,GST,GSH was found fallen and MDA increased in pathological group compared with control group (P<0.01). Resveratrol can raise the enzyme activity and reduce the level of MDA[Conclusions] This research shows that resveratrol can improvement the disorder of blood lipid and relieve the plaque. It also can enhance the activity of GSH-PX,CAT,T-SOD,GST,GSH, while decrease the level of MDA in a does-dependent manner; thus the progress of AS could be partially inhibited. Anti-oxdative stress may be one possible mechanism of resveratrol in anti-atherosclerosis effect.
