Objective:Hypertriglyceridemia is one of the important manifestations of dyslipidemia.Studies have shown that the level of triglycerides in human plasma is a risk factor for cardiovascular and cerebrovascular diseases,It is closely related to the occurrence and development of diabetes.In this study,we established a relatively stable and reasonable model of hypertriglyceridemia in SD rats,The consistency of the animal model was verified by the experimental results in and between laboratories,The factors that affect the blood lipid of rats were analyzed and compared.The focus of the experiment is to observe the changes of blood lipid.Methods:According to different types of animal models of dyslipidemia in SD rats,the general effects of different types of high-fat diets on serum lipids in rats were analyzed.To compare the effects of hydrogenated coconut oil and butter on TC and TG in rat plasma.After the high TG model was established with different concentrations of tallow feed,the expression of TC and TG in rat plasma and its influencing factors were observed.The results of high TG model were prepared in laboratory and laboratory.Variance analysis was performed first,and then the repeatability and consistency of the animal model and its influencing factors were discussedResults:High TC and hyperlipidemia animal models can be established repeatedly by using high TC and two-item model feed.When 15%lard feed was used for modeling,it took longer time to raise plasma TG than other modelsHigh TG models were built with hydrogenated coconut oil and 10%butter.When comparing their modeling effects,the tallow high-fat feed can rapidly increase the plasma TG level of rats,and maintain the TC and LDL-C levels relatively stable.It is better than hydrogenated coconut oil feed,and is an ideal feed for high TG modelWhen the high-triglyceride model was established with 10%tallow feed,the plasma TG concentration in the rats was 2.4 times higher than that in week 0 and nearly 4 times higher than that in the control group,which was 3.5 times higher than that in the control group.During the modeling process,plasma TC in rats was slightly increased,not more than 22%higher than the initial TC,and not more than 27%higher than the corresponding control group.The results of variance analysis showed that there was no time interaction between the concentration of tallow in feed and TC in rat plasma.When correcting rats' food intake and initial TC concentration,the effect of butter concentration in feed on TC in plasma was not statistically significant.There was no significant change in TC in the modelThe results of variance analysis in laboratory and inter-laboratory experiments showed that there were no statistical differences in body weight and blood lipid in rats The overall difference is due to differences in indicators between laboratories.There was no interaction between laboratory and laboratory replicationsConclusion:1.During the preparation of animal models with dyslipidemia,the animal model with high TC and hyperlipidemia SD had good repeatability and consistency.2.A high sugar and fat diet containing 10%butter,20%sucrose and 10%fructose rapidly raised the plasma triglycerides of SD rats3.The animal model of high triglyceride was successfully established by using high-sugar and high-fat feed containing 10%butter.In the model rats,plasma TG increased by 298%,and stabilized for 10 weeks4.Validation within and between laboratories,The establishment method of the model has good repeatability and consistency when the experiment is repeated in the laboratory.The difference between laboratories is due to the initial state of the experimental animals.
