Objective:To establish rabbits model of atherosclerosis. To investigate the effects and the mechanism of resveratrol on anti-inflammatory and anti-atherosclerosis effects in experimental atherosclerosis rabbits.Methods:Sixty male Japanese white rabbits were randomly divided into 6 groups(10 rabbits for each group):group A (normal control group) fed with normal feedstuff group B fed with high cholesterol feedstuff 10 weeks; group C fed with high cholesterol feedstuff 20 weeks; group D (treated with low dosage resveratrol 5mg/kg·d), group E (treated with mid dosage resveratrol 15mg/kg·d) and group F (treated with high dosage resveratrol 45 mg/kg·d) fed high cholesterol feedstuff combined with resveratrol. Only group B was fed for 10 weeks, the rest groups were 20 weeks. From the end of the 10th week, the rabbits was feeding accompanied by oral administration of resveratrol for ten weeks. Serum total cholesterol (TC), triglyceride (TG), low density lipoprotein-cholesterol (LDL-C) and high density lipoprotein-cholesterol (HDL-C) of the rabbits were detected by biochemical method at the first day and the end of the 20th week. At the end of the 20th week, animal were killed, the hearts and arteries were taken to do the Hematoxylin-Eosin (HE) staining. The level of TNF-α,IL-1βand IL-6 in serum were determined. The data was analyzed by the SPSS software package and the value of P<0.05 was statistically considered significant.Results:1. The atherosclerosis model with rabbits had been successfully established with high fat diet at the end of 10th week. There is no AS plaque found in group A. The terrible AS pathological changes can be found in the other groups. After treated with resveratrol, the lesion improved. The effect in group E and F are more significant than group D.2. After established the animal model, the blood lipid level of group B and group C significantly increased compared with control group(group A) (P<0.01, P<0.01); After treated with low does resveratrol, the level of TC,TG,LDL-C decreased compared with pathological group (P<0.01), otherwise the level of HDL-C incresed. The effect in group E and F are more significant than group D.3. The level of inflammatory cytokine was found increased in pathological group(group B and group C) compared with group A (P<0.01, P<0.01). After treated with resveratrol, the level of TNF-α,IL-1βand IL-6 decreased compared with group C. Resveratrol can inhibit inflammatory process by reduce the level of TNF-α,IL-1βand IL-6.Conclusions:1. The atherosclerosis model with rabbits had been successfully established with high fat diet at the end of 10th week.2. Resveratrol can improvement the disorder of blood lipid, after treated with resveratrol, the level of TC,TG,LDL-C decreased, otherwise the level of HDL-C incresed; Resveratrol can maintain endothelial function and relieve the square of plaque, thus the progress of AS could be partially inhibited.3. The activity of anti-inflammatory of resveratrol:resveratrol can reduce the level of TNF-α,IL-1βand IL-6.
