Objective:Observe the effects of Fuzhengxiezhuobaoshen decoction on 5/6 nephrectomized CRF rat model and discuss parts of the mechanisms of this prescription in the treatment of CRF.Method:Establish CRF model using 5/6 nephrectomy. Randomly put 60 SD male rats into 6 groups, namely, sham operation group, model group, control group, and three other groups with large, medium and small doses. Fuzhengxiezhuobaoshen decoction is administrated at 29.4g/kg, 19.6 g/kg and 9.8g/kg per body weight by gavage in the large, medium and small-dose groups respectively. Normal saline is administrated by gavage in sham operation group and model group. Kill the rats 8 weeks after the administration, measure the amount of plasma leptin(LP) via RIA, blood routine, and renal function, and observe renal pathological change.Results:RIA and renal function test results: No noticeable abnormal LP content and Scr,BUN level are detected in the sham operation group. Higher in model group and treatment groups than in sham operation group(p﹤0.05). Markedly lower in all treatment groups than in model group(p﹤0.05). Marked reduction is observed in the large and medium dose groups, but no apparent difference is noted between the two groups(p>0.05).No significant difference is observed between the small-dose group and Niaoduqing group.Blood routine test results: No noticeable abnormal RBC and Hb levels are detected in the sham operation group. Lower in model group and treatment groups than in sham operation group(p﹤0.05). Markedly higher in all treatment groups than in model group(p﹤0.05). Marked increase is observed in the large and medium dose groups, but no apparent difference is noted between the two groups(p>0.05). No significant difference is observed between the small-dose group and Niaoduqing group.Observation on renal pathology: Normal in the sham operation group. Different degrees of glomerular size enlargement, glomerular mesangial cell and matrix hyperplasia,glomerulosclerosis,renaltubular atrophy, and renal interstitial fibrosis are observed in model group and all treatment groups. Lower degrees of lesions are observed in all treatment groups than in model group, notably in the large and medium dose groups.Conclusions:1.The study shows that by reducing the content of LP, Fuzhengxiezhuobaoshen decoction improves CRF energy metabolism disorder, reduces renal pathological damage and delays CRF development and exacerbation.2. Fuzhengxiezhuobaoshen decoction in medium dose is the effective dosage for the treatment of CRF.
