Objective:To study the effects of Bushenqudu Decoction on chronic renal failure(CRF) patients in early and medium stage with serum creatinine (Scr), blood urea nitrogen (BUN), Systatin C, endogenous creatinine clearance rate (CCr) and the expression of renal transforming growth factor-β1 (TGF-β1), hepatocyte growth factor (HGF) in CRF rats modeled by adenine, in order to explore the apeutic effects and mechanism of Bushenqudu Decoction of curing CRF in early and medium stage.METHODS:1. Clinical observation:To collect 64 patients with chronic renal failure in early and medium stage and randomly divided them into control group and treatment group. Based on the regular therapy, the two groups of patients were given Niaoduqing particles or Bushenqudu Decoction on purpose. These treatments last for 2 consecutive months. Both before and after these treatment, the patients were examined with Scr, BUN, Systatin C, CCr. 2. Animal experiment:80 male SD rats were randomly divided into control group, model group, Niaoduqing High dose group, Niaoduqing middle dose group, Niaoduqing low dose group, kidney Bushenqudu Decoction high dose group, Bushenqudu Decoction middle dose group and Bushenqudu Decoction low dose group. Irrigation adenine and administration progressed in the same day. model making last for 30 days, and administration last for 35 days. After that, rats blood samples were collected through abdominal aortic to examine Scr, BUN. And kidney tissues samples were collected for determination of the expression of TGF-β1, HGF by immunohistochemistry method.Result:1. Clinical observation:Compared with the two groups'before these treatment, Scr,BUN,Systatin C of the two groups after that were significantly decreased, CCr increased (P<0.05). And the treatment group is more effective than the control group. There were statistically significant differences between the two groups.2. Animal experiments:①BUN,SCr:Comparing with the control group, the expression of BUN,SCrin all the others'were significantly increased(P<0.01). Comparing with model group, the expression of renal BUN,SCr in each group of Niaoduqing Particles and Bushenqudu Decoction increased significantly; And each group of Bushenqudu Decoction were more effective than Niaoduqing Particles in the same dose(P<0.01 or P<0.05);②TGF-p1:Compared with the control group, the expression of TGF-β1in all the others'were significantly lowered(P<0.01). Comparing with the model group, the expression of renal TGF-β1 in each group of Niaoduqing Particles and Bushenqudu Decoction decreased significantly; And each group of Bushenqudu Decoction were more effective than Niaoduqing Particles in the same dose(P<0.01 or P<0.05);③HGF: Compared with the control group, the expression of HGF in all the others' were significantly increased. Comparing with model group the expression of renal HGF in each group of Niaoduqing Particles and Bushenqudu Decoction increased significantly; And each group of Bushenqudu Decoction were more effective than Niaoduqing Particles in the same dose(P <0.01 or P<0.05); But among each group of Niaoduqing Particles or Bushenqudu Decoction, the expression of BUN,SCr and renal TGF-β1, HGF showed no significant difference neither.Conclusions:Both of Bushenqudu Decoction and Niaoduqing particles can significantly improve the function of CRF patients in early and medium stage, which is also able to promote the expression of TGF-β1 and inhibit the expression of HGF in the renal issue of CRF rats model. And the effect of Bushenqudu Decoction is better than Niaoduqing particles. The fact above indicates that Bushenqudu Decoction has the ability of improving renal function, slowing down the progress of renal function. The effect of promoting the expression of TGF-β1 and inhibiting the expression of HGF may be one of the one of the mechanisms. It is necessary for ur to take more attention on the therapeutic effect of the CRF and make a further promote research.
