| Objective:Based on the Polycystic Ovary (PCO) rat model built from Letrozole, researches are done to the effect of the law and its group of kidney Huayu Recipe on PCOS serum hormones and ovarian morphology to explore how the bu shen hua yu decoction PCOS affects hormone in rats and morphological mechanism and provide a theoretical basis for the clinical treatment of PCOS.Methods:In this experiment, according to the pre-experimental modeling results,Letrozole was chosen to create animal models.79 6-week-old SPF SD female rats with regular estrous were randomly divided into model control group,kidney Huayu Recipe on high, medium and low-dose group,positive medicine control group and blank control group.After drug intervention,serum hormone levels of the rats were examed and morphological changes of ovary were observed to explore the related mechanism of how the kidney Hua yu Recipe treat PCOS.Results:1.The evaluation of the success of animal models:According to the pre-experimental evaluation results,the reference to the reseach of Hu Yan,Zhang Zhan and Jia Li-ting about Letrozole inducing rat model of PCOS which is from the 957th-959th content of the 21th China's Maternal and Child Health published in2006 and the result of the experimental evaluation,the establishment of animal model of PCOS is determined through the measure to vaginal exfoliated cells,ovarian morphology and sex hormones,insulin.2.Determination of serum hormone levels:After being modeled, FSH,P, PRL,T levels did not change significantly, while the LH, INS levels went up and the E2 levels went down.After being poured into stomach with Kidney Huayu Recipe,the INS level of the high, medium and low-dose group was obviously lower than that of model group, the LH level of the low-dose group and the positive treatment group significantly decreased compared with the model group while there is no significant difference between the E2 level of high, medium and low-dose group and model group but the positive treatment group was significantly different from model group.3.Ovary Morphology:light microscopy results:Blank control group shows a large number of yellow bodies and follicles of different developmental stages.Those follicles were developing normally with its granulosa cell being dense and theca cells being spindle-shaped arranged.Blood group was visible in some ovaries.A small number of yellow cells,a large number of atretic follicles and the primordial follicle were found in model group with follicle cell layer thickening, incompletely developed follicle and the reduction of granule cell layer.The modeling was proved successfully according to the above.Comparing the Chinese medicine,high, medium and low-dose group, low dose group had the best approach with a lot of yellow bodies seen through the microscope which were more than high, medium-dose group.Besides,a large number of normal follicles, some secondary follicles and mature follicle and a small amount of follicular atresiar can be seen in it and the layers of granulosa cells increased.Compared with the positive drug, three dose groups of Chinese medicine was less effetive.A large number of yellow body,a lot of follicles and the follicles at different developmental stage can be seen in the positive drug through microscope while few atretic follicles can be found.Ganulosa cell layer in the positive drug increased in normal morphology which stood in neat row and follicular cell membrane became thin.Conclusion:1 the PCOS animal model is made successfully;2 The Kidney Huayu Recipe has the function of improving the PCOS big mouse internal secretion condition by increasing the PCOS big mouse's ovary granular cell and the yellow bodies,turning the graafian follicle cell membrane thin and obviously lowing blood serum INS,the LH hormone level;3 The Kidney Huayu Recipe can improve the internal secretion of the PCOS and the ovulation barrier condition through its.adjustment to the cerebral ganglion pituitary gland-ovary axis and the cerebral ganglion-pituitary gland-adrenal gland axis.
