| Backgrounds and purposeMigraine is a common headache.The clinical manifestations of migraine is a paroxysmal side or bilateral/pulsatile pain was moderate to severe pain?Migraine duration of about 4 to 72 hours,daily activities can aggravate headache,and often accompanied by nausea and vomiting,photophobia,phonophobia and other symptoms of autonomic dysfunction.may have a family history.The prevalence rate of migraine was 8.4 to 28%in the world.Long term repeated attacks of migraine seriously affect the mood of patients,affect family harmony,but also reduce the quality of life and work efficiency,to the social economy has also brought a huge loss.Migraine is a risk factor for cardiovascular and cerebrovascular diseases.Migraine may be associated with depression,bronchial asthma,epilepsy,and so on.At present,the pathogenesis of migraine is not well understood,and there is no special preventive treatment.Although the efficacy of Western medicine in relieving symptoms of acute attack of migraine has been recognized by evidence-based medicine,but there are many adverse reactions,such as dependence and addiction,which can lead to drug overuse headache.Preventive drugs often limit their use because of poor drug tolerance or past medical history.A number of studies show that the effect of traditional Chinese medicine treatment of migraine is no less than western medicine.At present,more and more people pay attention to the traditional Chinese medicine treatment of migraine.In this experiment,through the establishment of migraine rat model,observe the effect of Huoxue Shufeng Granule on the expression of plasma CGRP,SP content and c-fos of brain stem of rats,to explore the possible mechanism of activating blood and expelling wind particles in migraine prevention.MethodWistar rats were randomly divided into 6 groups,each group had 10 rats:normal control group,model group,flunarizine group and Huoxue Shufeng granules in high,medium and low dose group.Normal control group and model group were given the same volume of normal saline,the other groups were given the corresponding medicine,2 times a day,continuous gavage for 7 days.After seventh days,except for the normal control group,the rats in the control group were injected with normal saline,and the other groups were injected subcutaneously with Nitroglycerin Injection.Observe the behavior of rats:Red ear and scratching the appearance and disappearance of the time,the number of head.The rats were sacrificed after observing the behavior,and the plasma and brain stem tissues were taken.Plasma SP and CGRP levels were measured by enzyme-linked immunosorbent assay.The expression of c-fos in brain stem was determined by immunohistochemistry.Result1.The effect of granule for activating blood circulation and dispersing wind on the behavior of rats:Compared with the model group,Huoxue Shufeng granules in high and medium dose group and flunarizine significantly prolong redness both ears and forelimb scratching appears,and redness both ears and forelimb scratching disappeared time was shorter(P<0.05).Huoxue Shufeng granules in high and medium dose group and flunarizine group rats significantly reduced the number of forelimb scratching in each time period(P<0.05).2.The effect of Huoxue Shufeng granules on the content of SP and CGRP and the expression of c-fos in brain stem of rats:Compared with normal control group,the content of SP and CGRP and the expression of c-fos in the brain of model group were significantly increased(P<0.05).Compared with the model group,Plasma SP and CGRP content and c-fos expression in brainstem were significantly reduced in Huoxue Shufeng granules in high,medium and low dose group and flunarizine group(P<0.05).ConclusionThe experimental results show that,Huoxue Shufeng granules can improve the behavior of rats with migraine induced by nitroglycerin,and also can reduce the content of SP and CGRP in the plasma and decrease the expression of c-fos in the brain stem.Therefore,reducing the release of SP,CGRP and decreasing the expression of c-fos may be one of the mechanisms of the effect of Huoxue Shufeng granules on migraine. |
