Objective:Through the detection of the expression level of p-p38MAPK in pulpy nucleus of human degenerated lumbar intervertebral discs, to investigate the relationship of P-p38MAPK in the development of Lumber Disc Herniation(LDH),and to understand the pathology of Lumber Disc Herniation(LDH).This may provide an experimented proof for the treatment of clinical Lumber Disc Herniation(LDH).Methods:Hemiated lumbar disc tissues were obtained from 54 Patients under going surgery for Lumber Disc Herniation(LDH). Ages of the subjects ranged from 34 to 53 years, with 30 males and 24 females. All samples were from patients in The First Hospital of Shanxi Medical University from July,2008 to January,2010.16 control samples were the intervertebral discs from patients suffering from idiopathic scoliosis. Ages of the subjects ranged from 11 to 21 years, with 7 males and 9 females. The fresh pulpy nucleus tissue was obtained surgically and were fixed in 10% formalin, imbedded in Paraffin and sliced by 4μm. The degeneration of pulpy nucleus tissue was observed with HE staining and detected by means of immunohistochemical staining. The positive P-p38MAPK cells contain yellow or brown grains when observed under microscope. We choose 10 fields of view in random, then count masculine cells. The results are analyzed by statistics.Results:1.Histopathologic examination of herniated intervertebral disc tissues showed granulation tissue at the peripheral portions of herniated discs.2. The results of Evision staining:p-p38 protein expresses in nucleus, there were 48 cases of positive expression of p38 protein in the experimental group, Positive cell percentage is 88.89%(48/54), while positive cell percentage in the control group is 12.50%(2/16). The contrast has statistical significance(P<0.05).3. There is no correlation between the content of P-p38MAPK and the kind of hernia (P>0.05).Conclusions: 1. The quantity of P-p38MAPK increased in degenerative lumbar intervertabral disc. P-p38MAPK were involved in the pathogenesis of lumbar disc degeneration and lumbar disc hernaiton.2. There is no correlation between the content of P-p38MAPK and the kind of hernia.
