Objective 1.To study the correlation between MRI score of responsible focuses and ODI index in LDH.2.To study the role of AQP1,3 in the process of electrophoresis of the model rabbit in the degenerative lumbar intervertebral disc,to explore the correlation between water channel protein(AQPs)and the value of ADC value of intervertebral disc water dispersion. Methods 1.Clinical research: Among the 60 patients who were eligible for inclusion criteria, the choice accords with the symptoms and signs the segment and the level of use of nerve root compression Disc holder rate index of two images by MRI ratings, reuse ODI to score of patients with symptoms and signs, and finally to MRI and ODI scores for statistical analysis.2.Experimental study: 50 healthy adult male New Zealand white rabbits were selected and randomly divided into five groups: the normal group model group, the electroacupuncture group, the electroacupuncture group and the blocking agent group, each group of 10,adaptive feeding for three weeks.Normal group of conventional breeding is not processing, model group the rabbit model of intervertebral disc degeneration using homemade pressurizer, false model group with model group, but not on pressure between vertebral body, electric acupuncture group after the success of the building shall be cupping therapy, cupping plus blockers built after the success of the mold to the injection AQPs inhibitors after cupping treatment.Take 5experiment rabbit out of the first day and the 28 th day after the model cycle, then they were scanned by T2 WI,DWI and DTI.After processing the raw data of the scan, the pseudo-color map is generated to calculate the ADC and FA.The expression of water channel protein in the lumbar intervertebral disc tissue was detected by immunofluorescence assay. Results 1.Clinical research:According to the statistical analysis, the MRI score of the patient's responsibility lesion was correlated with the ODI index and showed a moderate positive correlation.2.Experimental study: When the value of the ADC value was 1d and 28 d after the model period, the normal group and the dummy model group were significantly higher than that in the model group and the electroacupuncture group(P<0.05).The ADC value and FA value were higher than the 1d after the model period, and the electroacupuncture group was significantly increased(P<0.05),while the electroacupuncture and blocker group did not increase the anti-drop(P<0.05).This shows that the experimental rabbit model of degenerative intervertebral disc production successfully, and cupping treatment improves the ADC FA values in degeneration, but the curative treatment and injected water channel protein inhibitors and reverse the effect of electric acupuncture therapy.Immunofluorescence assay showed the most positive expression of AQP1,AQP3 fluorescent protein in the normal group and the false model group, and the fluorescence brightness was evenly distributed. The fluorescence protein expression was the lowest in the model group, and the fluorescence attenuated the sparse electroacupuncture group was more positive than that in the model group.AQP1 showed a moderately positive linear relationship with FA,r=0.669,P<0.05.AQP1 has a direct correlation with ADC relative low level,r=0.613,P<0.05.AQP3 has a highly positive linear relationship with FA,r=0.874,P<0.05.AQP3 showed a moderately positive linear relationship with ADC,r=0.775,P<0.05.Conclusion 1.Clinical research: MRI score of lumbar intervertebral disc herniation and ODI index presented a positive correlation with the ODI index. The more serious MRI image was,the more serious the clinical symptoms might be.2.Experimental study: Cupping treatment can enhance the expression of AQP1 in degeneration and AQP3,adjust the water in the degeneration of intervertebral disc tissue metabolism, improve the dispersion ability of water molecules, the active role on the prevention and cure of intervertebral disc degeneration.ADC,FA andthe expression of AQP1,3 showed a linear correlation. It is very important to study the mechanism of intervertebral disc degeneration. |
