ObjectiveBy testing the expression of KiSSpeptin-54(Kp-54) and its receptor AXOR12 in villi and placenta tissue of different pregnant stages and severe preeclampsia, we analyze the different expression of Kp-54 and AXOR12, and then discuss the effect of Kp-54 and AXOR 12 in trophocyte invasion and the pathogenesis of preeclampsia.MethodsUse the immunohistochemistry to test the expression of Kp-54 and AXOR12 in the patients'villi and placenta tissue. The patients who are treated in China Medical University Shengjing Hospital during 2009.2-2009.12 are divided into two groups, one is the normal group (n=60), and the other is the preeclampsia group (n=40). The normal group divided anew into three subgroups, and they are the normal early pregnant subgroup (n=20), the normal medium-term pregnant subgroup (n=20) and the normal late pregnant subgroup (n=20).Result1. The expression of Kp-54 in the villi and placenta tissue is positive during the normal pregnancy, and the expression can be seen both in the cytoplasm of syncytiotrophoblasts and cytotrophoblasts. But in the syncytiotrophoblast cytoplasm, the positive expression is significantly stronger than that in the cytotrophoblasts', which is especially in the early stage.2. The strongest positive (+++) expression of Kp-54 which at the early pregnant stage is significantly higher than that in the medium-term pregnant stage and the late pregnant stage (P=0.002, P=0.002).3. The expression of AXOR12 which of the early stage and the mid stage cannot be detected, but in the late pregnant stage we find it, and it is mainly in the syncytiotrophoblast cytoplasm. 4. The weakest positive (+) expression of AXOR12 in the three subgroups have not statistically significance (P=0.152).5. During the normal pregnancy, the expression of Kp-54 and AXOR12 has no correlation.6. The location of the Kp-54 and AXOR12's expression in the severe preeclampsia group is similar to the normal late pregnant group, and their expressions are also mainly in the syncytiotrophoblast cytoplasm.7. The strongest positive (+++) expression of Kp-54 in severe preeclampsia group is significantly higher than that in the normal late pregnant group (P=0.027); but the expression of AXOR12 in the two groups has no significance (P=0.382).Conclusion1. This study shows that the expression of Kp-54 in normal early pregnant villi is significantly higher than that in the medium-term pregnant stage and the late pregnant stage, which suggest that Kp-54 may be involved in the regulation of the trophocyte infiltration and the development of the placenta, and it has an important effect for maintaining and continuing the pregnancy.2. During the normal pregnancy, the expression of Kp-54 and AXOR12 are not related, which indicate that the regulation of the trophocyte invasion by Kp-54 during the pregnancy may be not through its receptor AXOR12, it may be through other proteins or signal transducted pathway, and we should study further.3. The expression of Kp-54 in severe preeclampsia group is significantly higher than that in the normal late pregnant group, but the expression of AXOR12 in the two groups has no significance, these shows that Kp-54 may be involved in the pathogenesis of severe preeclampsia, and the effect of Kp-54 in the pathogenesis of preeclampsia is not through its receptor AXOR12, and we should study further.
