OBJECTIVETo investigate the expression of hypoxia-inducible factor-1α,Endoglin in placenta and serum soluble endoglin in Preeclampsia.To explore the role of HIF-1α,Endoglin and sEng in the Pathogenesis of Preeclampsia.METHOD1.Fourty women with Preeclampsia(including 10 mild and 30 severe Preeclampsia) and 20 normal pregnant women were recruited in department of obstetrics in Qilu hospital of Shandong university,according to the diagnostic code on the textbook《obstetrics and gynecology》.The two groups were matched in maternal age and gestational age.All cases had no other complications of pregnancy and complications of internal medicines.2.The expression of HIF-1αand Endoglin in placenta collected from 40 pregnant women with Preeclampsia(including mild and severe Preeclampsia) and 20 normal pregnant women were determined by immunohistochemistry.3.The levels of soluble endoglin in serum was detected by enzyme linked immunosorbent assay.Determine the optical density,using a microplate reader set to 450 nm.Construct a standard curve by plotting the mean absorbance obtaine for each reference standard against its concentration,Using the mean absorbance value for each sample,determine the corresponding concentration of sEng from the standard curve.4.The data was analyzed by SPSS13.0 software wrap,statistical analysis of two group t-test and Pearson relation were performed.RESULTS1.HIF-1αwas mainly located in the cytoplasm or nucleus of placental villus syncytiotrophoblast,less in the interstitial cells.The expression of HIF-1αin preeclampsia placenta(0.273±0.068) was significantly higher than that in normal placenta(0.101±0.060;P<0.01),and was higher in severe group(0.294±0.053) than that in mild group(0.208±0.072;P<0.01).2.Endoglin was mainly located in the cytomembrane of placental villus syncytiotrophoblast.The expression of Endoglin in preeclampsia placenta (0.320±0.061) was significantly higher than that in normal placenta (0.189±0.046;P<0.01),and was higher in severe group(0.341±0.054) than that in mild group(0.265±0.046;P<0.01)3.The level of sEng in serum of the preeclampsia group(4.911±1.088ng/mL) was significantly higher than that of the controls(1.230±0.403ng/mL,P<0.01),and was higher in severe group(5.233±1.037ng/mL)than in mild group(3.940±0.526 ng/mL;p<0.01);4.Posotive correlation between the HIF-1αand Endoglin was found both in study group(r=0.68,p<0.01)and control group(r=0.85,p<0.01)5.Posotive correlation between the HIF-1αand sEng was found both in study group(r=0.86,p<0.01)and control group(r=0.89,p<0.01)6.Posotive correlation between Endoglin and sEng was found both in study group(r=0.41,p<0.01)and control group(r=0.78,p<0.01)CONCLUSION1.The high expression and location of HIF-1αprotein and Endoglin protein indicates that they are the key genes,which may induce the occurrence and development of preeclampsia.The expression of Endoglin can be induced by hypoxia and regulated by HIF-1α.2.Excess expression of Endoglin in placenta may induce higher level of sEng in serum and then participate the pathophysiological processes of preeclampsia.
