Objective:To investigate the expression of Tumor necrosis factor-α(TNF-α)and Interleukin-6(IL-6) in a rabbit model of atherosclerosis,and the effect of Xuezhikang.To explore the possible anti-atherogenic mechanism of Xuezhikang.Methods:40 healthy New Zealand rabbits were divided randomly into five groups: Atherosclerosis model group , feeded high fat diet freely;Xuezhikang group rabbits had intragastric with administration Xuezhikang 0.8g/Kg soluble in 2ml once a day;Xuezhikang and mevalonate group, rabbits had intragastric with administration Xuezhikang 0.8g/Kg and mevalonate150mg/Kg soluble in 2ml once a day, mevalonate group, rabbits had intragastric with administration mevalonate150mg/Kg soluble in 2ml once a day, rabbits in the four groups were underwent balloon-induced abdominal aortic wall injury and then fed the cholesterol-rich diet.sham-operated group, rabbits were fed standard chow. And then were fed for 8 weeks.Cholesterol (TC) and triglyceride (TG) and low-density lipoprotein cholesterol (LDL) in serum were detected ,and rabbits were killed to obtain aorta tissue ,then observe morpholog change and expression of TNF-αand IL-6 by immunohistochemistry .Results:37 rabbits completed the study ,and the other 3 rabbits died.As compared with the control group the level of TC, TG, LDL-C and the expression of TNF-αand IL-6 in rabbits model in the other four grups increased(P<0.01);Compared with the model group , the level of TC, TG, LDL-C in Xuezhikang group lower than that in Xuezhikang + mevalonate group and mevalonate group (P<0.05); The level of TC, TG, LDL-C and the expression of TNF-αand IL-6 in rabbits model group and Xuezhikang + mevalonate group and mevalonate group were not different significantly (P>0.05). Compared with the Xuezhikang + mevalonate group and mevalonate group the expression of TNF-αand IL-6 in rabbits model were different in the Xuezhikang group(P<0.01).While the expression of TNF-αand IL-6 in rabbits were not different between Xuezhikang + mevalonate group and mevalonate group(P>0.05). Conclusions (1)Xuezhikang can reduce the expression of TNF-αand IL-6 significantly.At the same time,xuezhikang had the effect on decreasing the serum levels of TC,TG ,and LDL-C significantly. (2)Xuezhikang had the effect on preventing the formation of the artery atherosclerosis Lipid and anti-inflammation mechanisms contribute to the beneficial effects of Xuezhikang in artery atherosclerosis formation.(3) TNF-αand IL-6 were risk factor of atherosclerosis which can be used as a parameter to predict AS risk.
