Objective Observation of Ligustrazine on cochlear blood flow in guinea pigs for clinical treatment of cochlear microcirculation disorders caused by disease and provide experimental basis.Methods The use of photochemical model law will be 20 health, external auditory canal and middle ear without abnormal white red head a successful model in guinea pigs were randomly divided into control group (sodium chloride) 5 and 15 experimental group. Experimental group was randomly divided into TMP5mg/kg,20mg/kg and 60mg/kg concentration groups of 5 guinea pigs. Liquid experimental group were external jugular vein intubation uniform input, per minute 2ul, continuing 30 minutes. Control group given injections of the same volume of sodium chloride disposal. The use of laser doppler flow meter to monitor the cochlear blood flow ,synchronization input into the computer for statistical analysis.Results TMP on the impact of live animals COBF experiment, all animals in the experiment rectal temperature controlled at 38±1℃, each dose group were not found dyspnea, cyanosis, edema, death phenomenon. COBF control group after administration of the injection period and observation period after discontinuation of treatment were maintained at relatively stable. TMP5mg/kg Group COBF at 5 minutes after the injection began to rise 20 minutes after the 35 percent rise, after a gradual decline, but still higher than baseline levels. Ligustrazine 20mg/kg group COBF after injection at the beginning of an increase of nearly 67%, injection process has been maintained in accordance with the level at which about 15 minutes after the injection of gradually declining, but still higher than baseline levels. Ligustrazine 60mg/kg group COBF after injection at the beginning of an increase of near 75%, started 10 minutes after injection decreased to higher than the base value of 45%, maintaining this level until after the injection period and the end of observation period. Using SPSS software, selection of single-factor analysis of variance (ANOVA) of experimental data for statistical treatment, different concentrations of drug group and control group COBF compared to the same time, there is statistically significant changes in blood flow (P <0.05), drug group and control group differences in changes in blood flow significantly. Using SPSS software, choose the level of the two factors that many duplicate measurements analysis of variance (ANOVA) of experimental data for statistical treatment,There is a difference in different concentrations of intravenous drug delivery group and control group(P<0.05) . COBF is different in different group, 40mg/kg group is stabler.Conclusions TMP on the regulation of cochlear microcirculation plays an important role.in different concentrations of TMP on cochlear microcirculation mechanism may be different.
