Objective:To establish the sciatic nerve compression model of rat which can control pressure and time simultaneous.To investigate the changes of neural blood flow when oppress with different pressures and sites in rat's sciatic nerve;To investigate the effect of rehabilitation training on neural microcirculation and functional recovery in rat with sciatic nerve compression.Methods:(1) The changes of neural blood flow were observed by oppressing the sciatic nerve with five grades pressures using Gasbag in 10 wistar rats.(2)The 20 rats were divided into two groups randomly-the distal compression group and the proximal compression group-and the changes of neural blood flow were observed in each groups.(3)The 20 rats were divided into two groups after making the model of sciatic nerve compression.The experimental rats swam everyday at the end of 1st week after operation.The control rats lived freely after operation.The rats in two groups were determined sciatic nerve function index, detected neural blood flow with Laser Doppler Flowmetry and taken to perform CD34 immunohistochemical,axon and myelin sheath staining at the end of 3rd week after operation.Results:(1) The neural blood flow changed significantly(P<0.01) when the sciatic nerve was oppressed with different pressures and it had the decreasing tendency with the pressures increasing;(2)The neural blood flow of each group decreased obviously(P<0.05).The effect of oppressing distal nerve on neural blood flow was more obvious than that of oppressing proximal nerve(P<0.01);(3)①The SFI of the swimming group was higher than that of the model group (P<0.01).②There was no significant difference in neural blood flow between the experimental group and control group(P>0.05).③CD34 immunohistochemical staining showed that the microvessel density in experimental group was higher than in control group(P<0.05).④Axon and myelin sheath staining:The thickness of myelin sheath,average numbers of myelinated nerve fiber per area and diameter of regenerating axon in experimental group were larger than those in control group(P<0.05).Conclusion:Mechanical compression can influence the neural blood flow obviously.The distal vessels of sciatic nerve are the major sources of nerve blood supply.The rehabilitation training can improve the function of the compressed nerve and promote the neural angiogenesis.
