Objective To provide the evidence for the application of DXS8378 and DXS6808 in forensic science and popμlation genetics by investigating their alleles and haplotype frequencies of the two X-STR locus in the Han popμlation lived in Shanxi province,and to discuss the possibility of applying them to forensic casework.Methods First of all,EDTA-blood specimens were collected from 252 unrelated individuals (164 females,98 males) in Shanxi Han population.Different tissues of one corpse were analyzed including blood,muscle,heart,liver and kidney et al.Samples of 20 family trios were investigated for discontinuity.Then All blood-specimens were anticoagμlated by EDTA,the Genomic DNA was extracted by phenol-chloroform method and the two locus DXS8378 and DXS6808 were amplified with PCR.The PCR products were typed on 8%non-denaturing polyacrylamide gel electrophoresis followed by 0.1%silver-staining.Lastly allele frequencies, genotype frequencies and haplotype frequencies were obtained by a direct calculation;test of Hardy-Weinberg equilibrium for female genotype data,comparing of allele frequencies between male and female population,and comparing of genetic data between different populations were performed by x~2-test;also genetic parameters were calculated.Results Among 164 females and 98 males unrelated individuals from ShanXi Han population, DXS8378 exhibited 5 alleles named 9~13 and core sequence is ctat,which gene sequence size is 192bp~208bp;DXS6808 exhibited 5 alleles named 5~9 and core sequence is tatc,which gene sequence size is 241bp~259bp.And DXS8378 detected 5 alleles named 9~13 and their frequencies ranging from 0.0286~0.4714,DXS6808 detected 5 alleles named 5~9 and their frequencies ranging from 0.0592~0.3244,allele frequencies differentiation was not observed between male and female populations.Respectively,11,10 genotypes of 2 X-STRs were detected in females,and both their genotype frequencies did not depart from Hardy-Weinberg equilibrium. Discrimination power for females of each 1℃were 0.7860,0.7643,probability of exclusion were 0.5495,0.52480.Total 20 haplotype were identified in males,the gene diversity, discrimination power,and probability of exclusion were 0.9282,0.9888 and 0.9955,respectively. The typing results of different tissues from one individual were identical.No mutations were found from 20 family trios.Conclusion These results indicate that the DXS8378 and DXS6808 locus are high genetic polymorphic,the haplotype diversity shows good discriminating power and high excluding chance of paternity.They are high valuable in the application of forensic Medicine and population genetics.
