Objective To develop a set of new markers for forensic applications, D15S659 and D9S925 loci were chosen in present study. The polymorphism and allelic sequences of these loci were analyzed, and their forensic applications in Han population of eastern China are evaluated.Methods The EDTA-blood samples were collected from 109 unrelated individuals in Han population of eastern China. DNA was extracted with Chelex method. Each DNA sample was amplified using PCR for D15S659 and D9S925 loci. The PCR products were analyzed by non-denaturing vertical polyacrylaminde gel electrophoresis with continuous buffer system. The genetic and forensic parameters for the two STR loci were calculated by the Modified-Powerstats software.Results The allelic number of D15S659 and D9S925 was 8 and 6 respectively. The observed heterozygosity (H_o) of D15S659 was 0.826 and that of D9S925 was 0.798. The discrimination power (DP) of D15S659 was 0.944 and that of D9S925 was 0.894. The exclusion power (PE) of D15S659 and D9S925 were 0.648 and 0.596 respectively.Conclusion The distributions of allelic frequencies at two loci are satisfying in population studied. Present data suggest that D15S659 and D9S925 loci can be applied as new candidate markers for forensic individual identification and paternity testing.
