Objective To obtain the allele and haplotype frequencies of the two Y-STR loci(DYS622, DYS630)and discuss the possibility of applying them to forensic science by investigating these Y-STR loci in Han population of Shanxi.Methods Blood specimens were collected from 120 unrelated males in Han population of Shanxi to study the allele frequencies of the two Y-STR loci.The DNA was extracted by phenol/chloroform method.Different tissues of one healthy male corpse were analyzed including blood,muscle,liver,heart,brain and kidney.The blood of 20 females was collected as control. 15 father/son pairs were investigated.The alleles of DYS622 and DYS630 were amplified by PCR.The PCR products were analyzed on 8%non-denaturing polyacrylamide gel electrophoresis followed by silver-staining.Each allele of the DYS622 and DYS630 was sequenced and the allele ladders were constructed.Alleles of these Y-STR loci were nominated according to recommendations of the International Society of Forensic Genetics.The frequencies of these alleles were counted directly.Results In our study,six,seven alleles were found respectively in the DYS622 and DYS630 loci. The gene diversity of DYS622 and DYS630 is 0.7838,0.7844 respectively,and the discriminating power and excluding probability of paternity are the same.Among 120 Shanxi Han males,29 different haplotypes of these two loci were identified.This system has a haplotype diversity(HD)of 0.9552 and the discriminating power(DP)of 0.9552.Different tissue from one healthy male corpse also has the same allele in the two Y-STR loci.PCR products were not found in female DNA.No mutation was detected at the two loci by investigating the 15 father/son pairs.Conclusion These results indicate that the DYS622 and DYS630 loci are of high genetic polymorphism,the haplotype diversity shows good discriminating power and high excluding chance of paternity.They are to be useful for the test of male-female mixtures and paternity identification.
