Effect Of The Baicalin And Baicalin Zinc On Expression Of Candida Mycoderma Secretory Phospholipase And Antifungal Spectrum In Vitro

【Objective】1.To study two traditional Chinese medicines of Baicalin and Baicalin zinc antifungal activity on influence the antifungal spectrum of Candida albicans,Candida glabrata,Candida tropicalis,Candida parapsilosis and Candida krusei.2.To compare the activities of phospholipase of Candida mycoderma under the same condition.3.To discuss whether the fungistasis of the drugs on Candida have coincidence with the targets of the phospholipase in the base of comparing the quantity of phospholipase in Candida mycoderma between the drug-treated group and un-treated group.【Methods】1.To study the sensitiveness between Baicalin and Baicalin zinc through macroscopic observation,microscope quantitative method and MTT Microdilution method(according to the M27-A method defined by the National Committee for Clinical Laboratory Standards(NCCLS)). 2.five species of Candida mycoderma strains are cultured by the egg-yolk blate method at 37 degrees Centigrade,the diameters of the colony(A) and the colony plus precipitation zone(B) were measured,phospholipase activity was expressed with PZ=A/B,a low PZ means high phospholipase secretion.3.The quantity of phospholipase in Candida albicans between the drug-treated group and un-treated group is determined by liquid nitrogen grind and reverse transcription-PCR(RT-PCR).【Reslttlts】1.Candida albicans,Candida krusei,Candida glabrata,Candida tropicalis and Candida parapsilosis,which are the international standard Candida mycoderma,are sensitive to Baicalin and Baicalin zinc,MIC of Baicalin is 1mg/ml,3mg/ml,3mg/ml,1mg/ml,3mg/ml respectively;MIC of Baicalin zinc is 3mg/ml,3mg/ml,3mg/ml,1mg/ml,3mg/ml respectively.2.Candida mycoderma can produce different size white precipitation zone on the egg-yolk plate,PZ value at 37℃under the cμlture conditions 0.5, 0.96,0.93,0.78and lrespectively.3.To detect whether the inhibition of mRNA levels to inhibit the enzyme phospholipase synthesis,thereby inhibiting the generation of phospholipase,iquid nitrogen grind and reverse transcription-PCR (RT-PCR) indicated that there is obvious different between the drug-treated group and un-treated group.Baicalin,baicalin zinc with the MIC is 1mg/ml and 3mg/ml respectively,the un-treated group is completely inhibited.【Conclusions】1.Five species of Candida mycoderma whose MIC has a statistical significance are sensitive to Baicalin and Baicalin zic.2.Candida albicans can secretion a large number of phospholipase;The Candida krusei,Candida glabrata and Candida tropicalis secretion a small amount of phospholipase,while the Candida parapsilos is can not.3.Baicalin and Baicalin zinc have function of inhibiting phospholipase activities.This proves that two kinds of Chinese medicine to inhibit Candida phospholipase Synthesis in the level of mRNA.In a word,the phospholipase of Candida mycoderma secreting is one of the function target location of these two species of traditional Chinese medicine.
