| Objective To approach target site of baicalin against Candida albicans and s upply base for exploring antifungal action mechanism of the traditional Chin ese medicine active component by detecting MIC of baicalin for Candida al bicans and studying the effect of baicalin on succinate dehydrogenase, cytoc hrome-oxydase, Ca2+-Mg2+ATPase,cell cycle, apoptosis of Candida albicans th rough biochemistry and flow cytometryMethods 1 the Microdilution was applied to detect MIC of baicalin against Candida albicans and the effect of IgY on MIC of baicalin against Candidaalbicans according to M-27A method of NCCLS. 2 we used orthogonal des ign to detect the activity of SDH,CCO,Ca2+-Mg2+ATPase of Candida albic ans treated with baicalin, IgY. After incubated with different concentrations of baicalin and IgY for corresponding time, Candida albicans was harvested and resuspended in physiological saline, which was followed by freeze thawi ng and grind three times. Then chondriosome was extracted by differential c entrifugation and used as sample. Added the sample to corresponding agent with the enzyme,detected the rate of decay of absorbance by spectrophotome ter at 600nm,550 nm,640nm respectively to calculate the activity of SDH,C CO,Ca2+-Mg2+ ATPase. 3 Factorial design and flow cytometry were applied t o evaluate the effect of baicalin and/or IgY on cell cycle and Apoptosis of Candida albicans. Candida albicans treated with corresponding antifungal age nt for 12,24,48 hours respectively. was harvested and resuspended in PBS , which was followed by freeze thawing and grind several times,After staine d with PI at 4℃ for several hours, the mixture was filtered with 300 meshnylon net and sent to flow cytometry. |
