Objective:To observer the effect of recuperating-heart granule that it can to tonify Qi and nourishing the blood and stabilize the heart and calm the mind on rabbit model of similar unstable angina that the level of serum MCP-1,to discuss its mechanism of action for prevent and treat unstable angina,to provide a reliable theoretic basis for this therapy.Methods:28 rabbits,aortic wall injuries were introduced using an intravascular balloon in rabbits before they were fed on an a high fat diet for 8 weeks.At the end of week 8,according to the result of body surface ultrasonography,rabbits were randomly divided into five groups:there are model control group,recuperating-heart granule group,Cedocard group, atorvastation group and blank group.There are 7 rabbits in every group.The treatment groups take the drug for 14 days.At the end of 10 weeks,to copy the model of similar unstable angina that used the method of Pharmacological triggering.To observe the level of serum MCP-1 of the rabbits model through the method of ELISA.To investigate the thickness and blood flow rate of arteria carotis communis by body surface chromatic color ultrasonography and to observe pathomorphism change of blood vessel by HE dyeing.Results:1.The method of ELISA was used to exam the level of serum MCP-1 for rabbits,results show that compared with blank group,the level of serum MCP-1 obviously increase in model control group(P<0.01);Compared with model control group,the level of serum MCP-1 decreased obviously in each treatment group(P<0.05);Compared with Cedocard group and atorvastation group,the level of serum MCP-1 decreased obviously in recuperating-heart granule group(P<0.01).2.According to the results,of body surface ultrasonography,compared with the blank group,every group emerge that arteria carotis communis IMT obviously,thicken at the end of 8 weeks(P<0.01),RI and PI addition(P<0.01, P<0.05),but Vs of every group accelerate nubilously(P>0.05).At the end of 10 weeks,IMT,PI,RI of each treatment group decreased,compared with model control group,they are considered statistically significant(P<0.01, P<0.05),but in Cedocard group IMT descent obsoletly(P>0.05).IMT of recuperating-heart granule group and atorvastation group is lower than Cedocard group(P<0.05).But difference in recuperating-heart granule group and atorvastation group is nonsense(P>0.05).3.Pathology resules indicated,besides black control group,every group emerge that vascular endothelial cell damaged,endangium thicken,fatty deposition obviously,fatty substance plaque formed.Compared with the model group,each treatment group's arteria carotis communis endothelial cell decrease lightly,endomembrane thicken lightly,fatty deposition decrease under endomembrane,arterial media vascular smooth muscle cells proliferation,adventitia fibrosis and chronic inflammation cell infiltrated. Compared with the Cedocard group,Recuperating-heart granule group and Atorvastation group endomembrane fatty deposition are less than the Cedocard group,arterial media vascular smooth muscle arrange structuredlly fairly,adventitia fibrosis and chronic inflammation cell infiltrated lessen.Conclusion:Recuperating-heart granule can be significantly reduced the level of serum MCP-1 for rabbits.Recuperating-heart granule can be significantly improved abnormal atherosclerosis pathomorphology of the rabbit model,can restrain endangium hyperplasia.Recuperating-heart granule which can tonify Qi and nourishing the blood and stabilize the heart and calm the mind on the model of similar unstable angina has protective effect definitely.
