Experimental Study Of The Expression Of Transcription Factors OCT4,SOX2 In Gastric Carcinoma | | Posted on:2010-12-31 | Degree:Master | Type:Thesis | | Country:China | Candidate:M X Lin | Full Text:PDF | | GTID:2144360275475243 | Subject:Oncology | | Abstract/Summary: | PDF Full Text Request | | Objective:To investigate the significance and expression of transcription factors SOX2 and OCT4 in gastric carcinoma and the human gastric cancer cell line AGS being treated by dixitabine and to explore the mechanism of OCT4 and SOX2 gene in the occurrence and development of human gastric cancer and the potential value of treating gastric cancer with dixitabine.Methods:1.Immunohistochemistry was used to observe the expression with SOX2 and OCT4 in gastric cancer.2.Culture carcinomas cell line AGS,then detect the expression of SOX2 and OCT4 in the cell line being treated with dixitabine by immunohistochemistry and RT-PCR.In vitro,MTT method was used in the inhibition studies.Results:1. SOX2 expression level was lower in gastric cancer than that in normal gastriemucosa (P<0.05). SOX2 expression level showed positive expression of invasive depth(P<0.05);OCT4 expression level was higher in gastric cancer than that in normal gastriemucosa(P<0.05).OCT4 expression level was higher in undifferentiated carcinoma and poorly differentiated carcinoma than that in differentiated carcinoma and well-differentiated cancer(P<0.05).OCT4 expression level showed positive expression of invasive depth and lymph node metastasis(P<0.05).2. SOX2 and OCT4 expression were detected in gastric carcinomas cell line AGS;By RT-PCR and immunohistochemistry method ,it was confirmed that dixitabine could enhance the expression of SOX2 and it could inhibit the expression of OCT4 in the cell line;Dixitabine may be possess the potential value of treating gastric cancer .Conclusion:1. SOX2 expression level is lower in gastric cancer than that in normal gastriemucosa,it may be a tip that SOX2 lowly expression would relate to gastric glands cell malignant. OCT4 expression level increase by the degree of differentiation decreasing,OCT4 expression may maintain the poorly differentiated phenotype in gastric cancer.2. Dixitabine could enhance the expression of SOX2 and it could inhibit the expression of OCT4 in the cell line; Dixitabine could inhibit the proliferation of the gastric cancer cell line AGS.
