Objective: To explone the expression and significance of Sox2 and Oct4 in human glioma tissues of different pathological grades.Methods: Immunohistochemical technique was used to detect the expression of Sox2 and Oct4 on tissue microarray slice obtained from 45 cases of human glioma tissues and 5 cases of normal brain tissues.Results:(1)The immunopositive vatio of Sox2 was 2.61%±1.33 in normal human brain tissues,3.95%±3.40 in WHO Grade I,18.22 %±5.16 in WHO Grade Ⅱ,20.74%±5.14 in WHO Grade Ⅲ,60.15%±7.82 in WHO Grade Ⅳ,compare the expression of Sox2 in normal human brain tissues with human glioma tissues,respectively(F=87.9,P<0.01),it was significantly higher in grade Ⅲ- Ⅳgliomas than in grade I-Ⅱ,gliomas(P<0.01);(2)The Immunopositive vatio of Oct4 was 0 in normal human brain tissues,1.250%± 3.82 in WHO Grade I,37.20%±9.12 in WHO Grade Ⅱ,51.39%±9.72 in WHO Grade Ⅲ,63.15%±9.05 in WHO Grade Ⅳ,compare the expression of Oct4 in normal human brain tissues with human glioma tissues,respectively(F 值=67.7,P<0.01),it was significantly higher in grade Ⅲ- Ⅳgliomas than in grade I-Ⅱ,gliomas(P<0.01);(3)The relative expression level of Sox2 and Oct4 obviously correlated with the pathological grades,but there were not detected in normal human brain tissues(4)Sox2 expression in human glioma tissues was positively correlated with that of Oct4(r=0.619,P<0.01);(5)The expression of Sox2 and Oct4 were not related to patients age and sex(p>0.05).Conclusion: The expression of Sox2 and Oct4 in human glioma tissues in positively related to the malignant degree of the tumors.It is suggested that Sox2 and Oct4 may play an important role in the phatholgenesis and development of the human glioma tissues.Their high expressions in human glioma tissues may be one of the reasons of gliomas malignant and invasive growth.
