| Lonicera edulis is a kind of natural,green,wild edible berry.Lonicera edulis have the function of anti-inflammatory,decline blood pressure,antioxidant and anticancer ect,and is a hotspot at home and abroad.According to the principles of free radical damage,strong antioxidant capacity of the material may have potential anti-cancer function,so Lonicera edulis may become the future anti-cancer agents.Lonicera edulis was selected as an objective of study,and extraction technology of antioxidants was optimizated.We also studied the antioxidative activities of Lonicera edulis in vitro,and its scavenging activities of DPPH,·OH,O2-·and its anti-lard and anti-lipoperoxidation capitation,respectively.We carried on a thorough research about anti-cancer result and its synergistic function with ascorbic acid of extracts from Lonicera edulis.Using ultrasound-associated extraction,based on single factor experiment of ratio of liquid and raw material,supersonic time,supersonic temperature,supersonic power and alcohol content ratio,antioxidants of Lonicera edulis was extracted.Using response surface analysis extraction process was optimized,the DPPH as evaluation index,and this research established the optimum technological parameters of antioxidants.Optimal extraction conditions of antioxidants of Lonicera edulis were:ratio of liquid and raw material 26:1,supersonic temperature 52℃,supersonic power 160W,alcohol content 53%.Under the above mentioned conditions,the yield of the clearance effect on the DPPH was 67.13%.In order to investigate the antioxidative activities of Lonicera edulis in vitro,we studied its scavenging activities of DPPH,·OH,O2-·and its anti-lard and anti-lipoperoxidation capitation,compared with VC and VE.The results showed that extracts of Lonicera edulis possess remarkable scavenging effects against DPPH,·OH,O2-·in a dose dependent manner,and the scavenging capacities of the extract for·OH was higher than that of VC and VE.Anti-lard peroxidation of extracts was evidently,but its anti-lipoperoxidation was not as VC and VE.Compared to other extracts of fruit,inhibitions of extracts from Lonicera edulis.against the growth of human cancercells is better.Synergistic inhibition of extracts from Lonicera edulis.and ascorbic acid against the growth of HT29 cells was studied.The results showed ascorbic acid has no effect on the growth of HT29 cells,but ascorbic acid could increased the inhibition of extracts from Lonicera edulis.A typical DNA degradation associated with apoptosis was determined by DNA agarose electrophoresis.Typical DNA Ladder was very clear.The expression of several correlation apoptosis gene was analyzed.Extracts of Lonicera edulis didn't exert its antiapoptosis function through Bcl-2 and Bax,but it could promote apoptosis function of HT29 through activating Caspase-3 and Caspase-9,and could exert its antiapoptosis function through regulating antiapoptosis function.
